Abstract: RSARI and SUSTIPRIJATNO. Nitrogen is one of macroessential nutrients needed by rice. Half of nitrogen applied to plants lost due to evaporation, denitrification, and leaching. CsNitr1-L is a gene encodes nitrite transporter in plants, which transfers nitrite from cytosol into chloroplast. This research has been done to measure expression rate from CsNitr1-L gene on F2B3 transgenic rice and to determine optimum fertilizer amount. Research started by phenotype selection using 20 mM KNO2. DNA from rice leaves amplified using PCR with 35 S primer. Plants contained CsNitr1-L gene was fertilized summed equal with 0, 100, 200, and 300 kg/ha on field, respectively. Total RNA from rice leaves was isolated and was reversetranscriptased to cDNA. CsNitr1-L gene expression was analysed using Real Time PCR with actin as reference and unfertilized nontransgenic rice as calibrator. Result shown that transgenic rice with 100, 200,and 300 kg/ha fertilization, respectively, had gene expression up to 42-50 times compared to calibrator. While transgenic rice without fertilization had gene expression up to 99 times compared to calibrator. Optimum amount of fertilizer to stimulate pinnacle gene expression was found out to be around 0-100 kg/ha. Gene expression of undetected shows nitrite transporter gene was inactive.
