dc.description.abstract | The number of people who has been infected by HIV AIDS in Indonesia increases year to year. Antiviral drug development research is absolutely needed. Therefore, it’s important to develop a virus model that has similar characteristics to considering the use of the HIV virus as a model because it has a higher security risk. This research is the first step for the development of HIV model virus from Simian Retrovirus (SRV) Indonesian isolate based on specific characteristics are obtained. The purpose of this study was to perform, to isolate and to identify the pol Simian Retrovirus (SRV) gene from Macaca fascicularis and Macaca nemestrina which can then be characterized its nucleotide sequence, filogenetic analysis, amino acid sequence, conserve domain, and the modeling of three-dimensional structure using bioinformatics analysis. Isolation performed by PCR using a pair of primers 3284U and 4925L. Amplification of pol gene by PCR produced specific bands 1600 bp. The analysis showed that viral DNA sequences present in the samples of Macaca fascicularis is SRV 2 while the virus present in the samples of Macaca nemestrina is Simian endogenous retrovirus. Therefore, the three dimensional structure of SRV virus from Macaca nemestrina can not be analyzed. The results of the isolation and structural modeling of protein-coding genes reverse transcriptase and RNase H SRV-2 from Macaca fascicularis Indonesia has been successfully performed. Active side of the two enzyme has also been identified and determined its amino acid sequence. The Active side of RNase-H consists of three amino acids and reverse transcriptase consists of 27 amino acids. The filogenetic analysis of SRV-2 enzyme showed that reverse transcriptase and RNase H from Indonesian isolates can be used as a model to study the enzyme from other retroviruses. | en |
dc.description.abstract | Jumlah penderita HIV AIDS di Indonesia meningkat dari tahun ke tahun. Penelitian pengembangan obat antiviral mutlak diperlukan. Untuk itu, perlu dikembangkan model virus yang memiliki karakteristik mirip dengan HIV mengingat penggunaan virus HIV sebagai virus model memiliki resiko keamanan yang lebih tinggi. Penelitian ini merupakan langkah awal untuk pengembangan virus model HIV dari Simian retrovirus (SRV) isolat Indonesia berdasarkan karakter-karakter spesifiknya. Tujuan penelitian ini adalah melakukan isolasi dan identifikasi gen pol Simian Retrovirus (SRV) isolat Indonesia asal Macaca fascicularis dan Macaca nemestrina yang selanjutnya dapat dikarakterisasi urutan nukleotidanya, dianalisis kekerabatannya, urutan asam aminonya, conserve domainnya, dan pemodelan struktur tiga dimensinya menggunakan analisis bioinformatika. Isolasi dilakukan dengan teknik PCR menggunakan sepasang primer 3284U dan 4925L. Amplifikasi gen pol dengan teknik PCR menghasilkan pita spesifik yang masing-masing berukuran sekitar 1600 bp. Hasil analisis sekuens DNA menunjukkan bahwa virus yang terdapat di dalam sampel Macaca fascicularis adalah SRV 2 sedangkan virus yang terdapat di dalam sampel Macaca nemestrina adalah Simian endogenous retrovirus. Oleh karena itu, virus SRV asal Macaca nemestrina tidak bisa dilakukan pemodelan struktur tiga dimensinya. Hasil isolasi dan permodelan struktur protein gen penyandi reverse transkriptase dan RNase-H SRV 2 asal Macaca fascicularis Indonesia telah berhasil dilakukan. Sisi aktif dari kedua enzim tersebut juga telah berhasil diidentifikasi dan ditentukan urutan asam aminonya. Sisi aktif dari RNase-H terdiri atas tiga asam amino dan reverse transkriptase terdiri atas 27 asam amino. Hubungan kekerabatan enzim dari SRV-2 isolat Indonesia menunjukkan bahwa enzim reverse transkriptase dan RNase-H isolat Indonesia dapat digunakan sebagai model untuk mempelajari enzim dari retrovirus lainnya. | |
