Abstract: Black seed (Nigella sativa) has been traditionally used as healing herbs worldwide. The aim of this study was to investigate the histopathological changes of female reproductive organs mice after treated by N. sativa oil extract. Thirty six adult female mice average four weeks age were divided into four groups, namely: Group I, per-orally (p.o) treated daily with 0.1 ml of aquadest; Group II, p.o treated daily with 0.1 ml of N. sativa extract; Group III, p.o treated daily with 0.2 ml of N. sativa extract; Group IV, p.o treated daily with combination of N. sativa extract and honey. After two months of treatment, necropsy was performed to collect ovaries and uterus of mice. Samples were routinely processed for histopathology, and stained with Hematoxylin-Eosin and Periodic-Acid Schiff. The histopathological parameters observed in this study were the size of ovary and its tertiary follicles, the number of developing follicles in ovary, the number of glands and goblet cells in uterus, and percentage of uterine re-epithelialization. As compared with the negative controls, N. sativa treatment increased the number of secondary follicles in ovary, initiated re-epithelialization in uterus. It is, therefore, considered that N. sativa may improve ovary performance and uterus homeostatic repair. This result would provide a baseline data for N. sativa effect in female reproduction system of mice.
Jintan hitam (Nigella sativa) secara tradisional digunakan sebagai obat herbal yang sedang popular. Tujuan penelitian kali ini adalah untuk mengetahui efek jintan hitam terhadap histopatologi organ reproduksi betina mencit. Mencit betina umur empat minggu sebanyak 36 ekor dibagi menjadi empat kelompok, yaitu kelompok I diberikan aquades 0,1 ml, kelompok II diberikan 0,1 ml ekstrak minyak jintan hitam, kelompok III diberikan 0,2 ml ekstrak minyak jintan hitam, dan kelompok IV diberikan kombinasi ekstrak minyak jintan hitam dengan madu sebanyak 0,3 ml. Mencit diberikan perlakuan satu kali sehari selama dua bulan. Setelah dua bulan perlakuan, dilakukan nekropsi untuk mendapatkan ovarium dan uterus. Selanjutnya organ diproses untuk histopatologi dengan pewarnaan Hematoksilin-Eosin dan Periodic Acid Schiff. Paramater yang diamati terdiri dari luas ovarium dan folikel tersier; jumlah folikel ovarium, kelenjar uterus, dan sel Goblet; dan regenerasi epitel permukaan uterus. Hasil yang didapatkan menunjukkan bahwa jintan hitam meningkatkan kinerja ovarium dan perbaikan homeostasis uterus. Kesimpulan dari penelitian ini yaitu jintan hitam memberikan efek terhadap sistem reproduksi betina mencit.
