dc.description.abstractWild Betta (Betta imbellis) is one of the Betta strain not cultured widely since body pigments are not so attractive. Color improvement of the fish may increase its attractiveness and can carried out by several methods, one of them is by transgenesis. The aims of this research are to evaluate the effectiveness of transfection method to deliver red fluorescent (RFP) gene into zygotes of B. imbellis and to observe its embryogenesis. Research was conducted in Balai Penelitian dan Pengembangan Budidaya Ikan Hias Depok and Balai Penelitian Pemuliaan Ikan Sukamandi, Subang for 6 months. The prospective broodstocks are selected and spawned in 1:1 ratio, followed by transfection of zygotes, DNA extraction, PCR, also the observation of embryogenesis. The treatments are RFP 1:1 and RFP 3:1 which repeated as much as 6 times. Hatching rate after transfection in each group did not show the clear pattern of treatment effect, but was not different significantly from non transgenic group overall. PCR from embryos and larvaes shown that DNA could be amplified in 0.6 kb DNA size. DNA bands also appear in non transgenic group so it has not been explain the success of transfection method to deliver fluorescent gene. The embryogenesis was taken place for approximately 34 hours after fertilization followed by hatching.en
dc.description.abstractIkan cupang alam (Betta imbellis) merupakan salah satu strain ikan cupang yang belum dibudidayakan secara luas karena warnanya kurang menarik. Usaha untuk meningkatkan kualitas warna pada ikan cupang alam salah satunya dapat dilakukan melalui transgenesis (transfer gen). Tujuan penelitian ini adalah melihat efektivitas metode transfeksi dalam penyisipan gen red fluorescent protein (RFP) pada zigot serta mengamati proses embriogenesis B. imbellis. Penelitian dilaksanakan di Balai Penelitian dan Pengembangan Budidaya Ikan Hias Depok serta Balai Penelitian Pemuliaan Ikan Sukamandi, Subang selama 6 bulan. Calon induk B. imbellis diseleksi dan dipijahkan dengan perbandingan 1:1, selajutnya dilakukan transfeksi telur, ekstraksi DNA, PCR, serta pengamatan embriogenesis. Perlakuan yang diberikan adalah RFP 1:1 dan RFP 3:1 masing-masing sebanyak 6 ulangan. Derajat penetasan telur setelah transfeksi tidak memperlihatkan pola yang jelas dari masing-masing perlakuan, namun secara umum tidak menunjukkan perbedaan yang signifikan dengan kontrol non transgenik. Hasil PCR pada embrio dan larva menunjukkan bahwa DNA teramplifikasi dengan ukuran sekitar 0.6 kb. Pita DNA juga muncul pada kontrol non transgenik sehingga belum dapat menjelaskan keberhasilan metode transfeksi dalam menyisipkan RFP ke genom B. imbellis. Embriogenesis berlangsung selama lebih kurang 34 jam setelah fertilisasi yang diikuti dengan penetasan
