Abstract: The separation of the active compound Sauropus androgynus leaf (SA leaf) based on their solubility of organic solvents (ethanol, hexan, ethyl acetate, and water). This research was conducted to elucidate the effects of the administration of SA leaf extract and fraction on the hematological views in lactating rats. The extract and fraction was mixed in the pelleted feeds by ratio of 0,87%, 4,53%, 0,45%, and 3,22% for hexan fraction (FH), extracts of ethanol (E-Eto), ethyl acetate fraction (F-EtOAc), and water fraction (F-H2O) respectively. Fifteen lactating rats were divided into 5 groups, such as control (K), FH , E-Eto, F-EtOAc, and F-H2O. The administration of SA leaf extract and fraction to each groups was conducted on the first pregnancy up to the 10 days lactation. After 10 days lactation period the measurement of the total erythrocytes and leucocytes, hemoglobin (Hb), hematocrit values (PCV), and the differential leucocytes was executed. The research revealed that the SA leaf extract and fraction administration showed no significant influence for all hematological parameters. But there was an indication the decreasing of the total leucocytes on F-EtOAc administration and also Hb values on FEtOAc and F-H administration. On the other hand, the F-H and E-Eto administration showed the increasing of the percentage of lymphocytes in leucocytes. The changes of hematological views, might be caused by the roles of active compounds in the extract and fraction of SA leaf.
Pemisahan kandungan senyawa aktif daun Sauropus androgynus (daun SA) dilakukan berdasarkan sifat kelarutannya pada pelarut organik (etanol, hexan, etil asetat, dan air). Penelitian ini dilakukan untuk menjelaskan pengaruh pemberian ekstrak dan fraksi daun SA terhadap gambaran hematologi pada tikus putih laktasi. Ekstrak dan fraksi daun SA dicampurkan ke dalam pelet dengan rasio 0,87%, 4,53%, 0,45%, dan 3,22% masing-masing untuk fraksi hexan (F-H), ekstrak etanol (E-Eto), fraksi etil asetat (F-EtOAc), dan fraksi air (F-H2O). Sebanyak 15 ekor tikus laktasi dibagi ke dalam 5 kelompok perlakuan, yaitu kelompok perlakuan kontrol (K), F-H, E-Eto, F-EtOAc, dan F-H2O. Pemberian ekstrak dan fraksi daun SA masing-masing kelompok perlakuan diberikan pada hari kebuntingan pertama sampai dengan laktasi hari ke-10. Setelah 10 hari masa laktasi dilakukan pengukuran terhadap jumlah eritrosit dan leukosit, kadar hemoglobin (Hb), nilai hematokrit (PCV), dan diferensial leukosit. Penelitian ini mengungkapkan bahwa pemberian ekstrak dan fraksi daun SA secara umum tidak mempengaruhi keseluruhan parameter hematologi. Namun ada tanda-tanda penurunan jumlah leukosit pada pemberian F-EtOAc dan juga penurunan kadar Hb pada pemberian F-EtOAc dan F-H. Selain itu, pemberian F-H dan E-Eto juga menunjukkan adanya peningkatan limfosit. Perubahan-perubahan yang terjadi pada gambaran hematologi tersebut kemungkinan disebabkan oleh peran senyawa aktif pada ekstrak dan fraksi daun SA.
