Abstract: Perkembangan jenis sapi di Indonesia berasal dari sapi asli seperti sapi bali dan juga sapi hasil silangan yang telah menjadi sapi lokal seperti sapi pesisir, sapi madura, sapi aceh, dan sapi Peranakan Ongole (PO). Sapi lokal Indonesia memiliki keanekaragaman tinggi seperti warna kulit maupun ukuran tubuh. Pendekatan penanda molekuler mampu membedakan bangsa sapi lokal Indonesia secara baik. Penelitian ini dilakukan untuk merekonstruksi pohon filogenetik beberapa bangsa sapi lokal Indonesia berdasarkan sekuens DNA mitokondria gen Cytochrome Oxidase I (COI). Sampel yang digunakan adalah sampel sapi bali, madura, aceh, pesisir, dan PO. Amplifikasi secara in vitro dilakukan dengan teknik Polymerase Chain reaction (PCR). 10 sampel berhasil diamplifikasi dan menghasilkan produk amplifikasi sebesar 710 pb, namun hanya delapan sampel yang berhasil dirunutkan dan dianalisis. Analisis hubungan kekerabatan pada sapi lokal Indonesia menunjukkan bahwa, sapi bali terpisah dari pengelompokan keempat bangsa sapi lainnya. Sapi aceh, pesisir, PO, dan madura terletak dalam kelompok yang sama dengan Bos indicus. Dari kedua pengelompokan tersebut menunjukkan bahwa sapi bali mempunyai asal-usul tersendiri sedangkan keempat sapi lainnya berasal dari kelompok sapi zebu.
The development of Indonesian cattle breeds derived from the original cattle such as bali cattle and also crossbreed which has become local cattle such as pesisir, madura, aceh, and filial ongole (PO) cattle. Indonesian local cattle has a high diversity such as color skin as well as body size. The approach of molecular markers can distinguish the Indonesian local cattle. This study was conducted to reconstruct the phylogenetic tree of several Indonesian local cattle based on mitochondrial DNA sequences of Cytochrome Oxidase I (COI) gene. The samples were used are bali, madura, aceh, pesisir, and PO cattle. In vitro amplification were done by using polymerase chain reaction (PCR) technique. 10 samples were successfully amplified and generated amplification products of 710 bp, but only eight samples were sequenced and analyzed succesfully. Phylogenetic analysis of Indonesian local cattle showed that bali cattle is separated from other cattles. Aceh, pesisir, madura dan PO cattle are located in the same group with Bos indicus. From these groupings showed that bali cattle have separate origins while the four others derived from zebu cattle group.
