Abstract: Giant gouramy is one of the targeted farmed species to increase its production level in the Indonesian Ministry of Maritime Affairs and Fisheries Program 2010-2014. Its production seemed slow because of the relatively long reproductive cycle. Transplantation of giant gouramy testicular cells to tilapia can be done in the hope that the surrogate broodstock (tilapia) will be able to give birth giant gouramy so the production cycle can be accelerated. In this research, transplantation of giant gouramy testicular cell (donor) in the peritoneal cavity of newly hatched tilapia (recipient) was performed. Giant gouramy testicular cells that contain spermatogonial cells obtained by dissociation of the gonad using PBS with 0.5% trypsin and then injected into tilapia with different ages. The results showed that SR (survival rate) of 1-2 days old injected larvae (A; 89.34%) were lower compared to that of 3-4 days old (B; 98.96%). Furthermore, the success rate of donor cells colonization in the recipient gonad at 2 months after injection in treatment A (6 of 6 recipients; 100%) was higher than that of treatment B (3 of 4 recipients; 75%). The results of this study indicated that transplantation using 1-2 days old larvae gave highest colonization and comparable level of SR. Thus, this technique has the potential both to manipulate gamet production of economically important fish species that relatively difficult to reproduction (commercial) and reproduce the endangered fish (as conservation).
