Deteksi Keberadaan Antibodi Anti-Escherichia coli di Dalam Serum Sapi Neonatus yang Diberi Kolostrum dengan Metode Enzyme-Linked Immunosorbent Assay (ELISA)

Abstract

Penelitian ini bertujuan untuk mengetahui gambaran antibodi di dalam serum sapi neonatus yang ditantang dengan Escherichia coli enterotoksigenik (ETEC) K99 sebelum dan setelah pemberian kolostrum. Sebelas ekor sapi neonatus jenis Friesian Holstein (FH) yang sehat secara klinis dipelihara sejak kelahiran sampai umur satu minggu. Sapi neonatus dikelompokan menjadi dua, yaitu kelompok kolostrum (8 ekor) dan kelompok non-kolostrum (3 ekor). Umur 0-72 jam, kelompok kolostrum diberi kolostrum dengan interval 12 jam, sedangkan kelompok non-kolostrum diberi susu sapi dengan waktu yang sama seperti kelompok kolostrum. Saat berumur 12 jam, semua kelompok perlakuan ditantang dengan ETEC K99 sebanyak 5X1010 CFU yang diemulsikan di dalam alhidrogel dan diberikan secara peroral. Pengambilan sampel darah dari vena jugularis dilakukan pada umur ke-0 (kontrol), 12, 24, 48, 72, dan 168 jam untuk mengetahui keberadaan antibodi di dalam serum dengan menggunakan uji ELISA (Enzyme Linked Immunosorbent Assay) tidak langsung. Hasil penelitian menunjukkan bahwa pada kelompok kolostrum, antibodi terdeteksi sejak 12 jam pemberian dengan konsentrasi tertinggi pada umur 24 jam, sedangkan pada kelompok non-kolostrum tidak terdeteksi adanya antibodi pada semua umur. Berdasarkan hasil penelitian dapat disimpulkan bahwa antibodi asal kolostrum dapat terdeteksi pada sapi neonatus yang diberi kolostrum sejak 12 jam sampai satu minggu setelah pemberian. Konsentrasi antibodi terhadap E. coli tertinggi adalah pada 24 jam setelah pemberian.

The aim of this research is to study the present of antibody against E. coli in the sera of neonates calf which given colostrum and challenged by enterotoxigenic Escherichia coli (ETEC) K99. Eleven neonates calves were clinically healthy kept since birth until the age of one week.This neonates calves are divided into two groups, first group is colostrums group (8 calves) and the other is non-colostrum group (3 calves). Colostrum group were given colostrum on age 0-72 hours with 12 hours interval, whereas non-colostrum group are given cow milk at the same time. Each group were challenge by ETEC K99 5X1010 CFU emulsion it in alhydrogel peroral at the 12 hours of age. Blood sample were taken from jugularis vein on age 0th, 12th, 24th, 48th, 72th, and 168th hours. The present of antibody in the sera detecting by indirect ELISA (Enzyme-Linked Immunosorbent Assay Method). The result showed of the highest concentration of antibody in the sera at 24 hours of age for colostrums group, whereas no antibody against E. coli were detected for non-colostrums group . Based on the results, we concluded the concentration of antibody in colostrum can detection at 24 hours until one week after given colostrum. The highest concentration of antibody anti- E. coli in 24 hours after given colostrum.
