dc.description.abstractGenomic library of soybean cv. Slamet had been constructed in λ phage vector. Partial digestion of total DNA was conducted using 0.014 units Sau3AI per μg DNA at 370C for 30 minutes. By using 0.5 μg insert DNA and the molarity ratio between insert DNA:vector = 2.9:1, the titer of 5 x 105 plaque forming unit was obtained. Among them, 85% or 4.25 x 105 pfu are recombinant. Since the length of insert DNA in λ recombinant phages is 15 kb in average, and the genome size of diploid soybean is 2.23 x 106 kb, therefore, the genomic library constructed was expected to contain all genome of soybean cv. Slamet.id
