dc.description.abstractTobacco Mosaic Virus (TMV) is one of important diseases of tobacco which sould reduce the yield and quality o f the crop. Mersitem culture could be applied on the production of virus free seedling. As a preliminary study, in this experiment explant was obtained from the apical meristem (including 2/3 leaf primordia) of healthy plant. Basal media used were M S + sucrose + vitamine (mesoinositol 0 -5 mg/l + thiamine HCl 0.1 mg/l, pyridoxine 0.5 mg/l and n i c o t i n a t e acid 0.5 m g / l) . The basal media were prepared in a liquid media enriched w i t h BA of 1, 3 and 5 mg/l, and in a solid media enriched with kinetin of 0.5; 0.1; 1.5; 2.0 mg/l. Rooting was induced with IRA and IBA at the concentration of 1, 3 and 5 mg/l respectively. Results showed that rooting in solid media was better than in liquid one. The media treated with BA the mereistem produced rosette + vitrification + calllus . Kinetin 0.5 mg/l was the best to use, producing 4 shoots w i t h 11.16 leaves per explant with in 4 weeks. To induce the elongation of roots the best medium was that enriched with IAA 1 mg/l. The growth of shoots was more vigorous on the media enriched with IAA than that enriched with IBA.id
