dc.description.abstractMastitis merupakan peradangan kelenjar ambing yang bersifat kompleks. Mastitis subklinis tidak menunjukkan perubahan pada ambing serta susu yang dihasilkan sehingga sulit untuk dideteksi. Namun, mastitis subklinis menyebabkan kerugian karena menyebabkan terjadinya penurunan produksi dan kualitas susu. Mastitis subklinis dapat dideteksi dengan melakukan monitoring jumlah sel somatik (JSS) yang hanya dapat dilakukan di laboratorium dan membutuhkan keahlian sehingga tidak praktis dilakukan di lapang. Penelitian ini bertujuan mengembangkan perangkat mikrofluida berbahan kain katun sebagai alat deteksi mastitis subklinis yang lebih sederhana, cepat, dan ekonomis. Matriks kain katun diberi perlakuan berbeda, dilapisi reagen California mastitis test (CMT), dan dipasangi conjugate pad untuk memfasilitasi pewarnaan sampel. Sampel susu sehat, mastitis subklinis, dan mastitis klinis diuji untuk mengamati pola rembesan plasma dan koagulan. Hasil penelitian ini menunjukkan bahwa kain katun yang dicuci dua kali memiliki laju perembesan yang paling baik, perbedaan JSS memengaruhi reaksi koagulasi serta jarak tempuh rembesan, di mana susu sehat memiliki jarak rembesan paling jauh dan mastitis subklinis paling pendek. Nilai Limit of Detection (LoD) dan Limit of Quantification (LoQ) menunjukkan kemampuan perangkat dalam mendeteksi variasi koagulasi pada sampel. Penelitian ini menyimpulkan bahwa perangkat mikrofluida berbahan kain katun dengan reagen CMT mampu mendeteksi mastitis subklinis dalam waktu 60 detik dan berpotensi digunakan sebagai alat diagnostik di lapangan yang praktis dan murah.-
dc.description.abstractMastitis is a complex inflammation of the mammary gland. Subclinical mastitis does not cause visible changes in the udder or in the milk produced, making it difficult to detect. However, subclinical mastitis resulting in economic losses due to reduced milk production and quality. Subclinical mastitis can be detected by monitoring somatic cell count (SCC) that can only be performed in the laboratory and requires technical expertise, making it impractical for field use. This study aimed to develop a cotton-based microfluidic device as a simpler, faster, and more economical tool for detecting subclinical mastitis. The cotton matrices were subjected to different treatments, coated with California Mastitis Test (CMT) reagent, and equipped with a conjugate pad to facilitate sample staining. Milk samples from healthy, subclinical and clinical mastitis cows were tested to observe plasma and coagulum wicking patterns. The results showed that the fabric that washed twice has the best permeation rate, the differences in SCC affected coagulation reactions and wicking distances, with healthy milk exhibiting the longest wicking distance and subclinical mastitis the shortest. The Limit of Detection (LoD) and Limit of Quantification (LoQ) demonstrated the device’s ability to detect variations in sample coagulation. This study concludes that the cotton-based microfluidic device with CMT reagent can detect subclinical mastitis within 60 seconds and has strong potential for use as a practical and low-cost field diagnostic tool.-
