Abstract: Rauvolfia serpentina merupakan tumbuhan herbal bernilai tinggi yang mengandung metabolit sekunder penting seperti reserpine, serpentine, dan ajmaline, namun populasinya kian menurun. Penelitian ini dilakukan untuk mengoptimalkan produksi biomassa R.serpentina demi mendapatkan senyawa-senyawa tersebut. Eksplan diperlakukan dengan konsentrasi pemadat Gelrite 1, 2, dan 3 g/L selama tiga bulan, dengan masing-masing 20 kali pengulangan. Observasi meliputi tinggi eksplan, jumlah daun, tunas, kalus, panjang akar, kontaminasi, dan browning. Hasil analisis (Uji F dan deskriptif kualitatif) menunjukkan bahwa empat dari lima parameter terdistribusi normal, namun pertumbuhan akar sangat minim. Parameter jumlah kalus menunjukkan pengaruh yang berbeda nyata. Perlakuan Gelrite 2 g/L terbukti dapat menumbuhkan kalus dengan optimal dibandingkan dengan perlakuan yang lain. Perlakuann Gelrite 1 g/L terbukti optimal karena mampu menyalurkan unsur hara dengan cepat, sehingga dapat memberikan hasil dari pertumbuhan parameter tinggi eksplan, jumlah daun, tunas, kalus, dan panjang akar dengan lebih optimal.
Rauvolfia serpentina is a valuable medicinal plant that produces essential secondary metabolites such as reserpine, serpentine, and ajmaline, but its population is declining. This study was conducted to optimize the biomass production of R. serpentina in order to obtain these compounds. Explants were treated with Gelrite concentrations of 1, 2, and 3 g/L for three months, with 20 replicates each. Observations included explant height, number of leaves, shoots, callus, root length, contamination, and browning. The analysist (F-test and qualitative descriptive) showed that four of the five parameters were normally distributed, but root growth was minimal. The number of callus parameters showed a significantly different effect. Gelrite 2 g/L proved to be able to grow callus optimally compared to other concentrations. Gelrite 1 g/L proved to be optimal as it distributed nutrients quickly, thus providing better results for the parameters of explant height, number of leaves, shoots, callus, and root length.
