Abstract: Padi (Oryza sativa L.) merupakan salah satu komoditas pangan utama yang memiliki peran strategis dalam mendukung ketahanan pangan nasional. Salah satu kendala dalam budi daya padi adalah infeksi nematoda parasit tanaman yang dapat menurunkan pertumbuhan dan hasil panen. Penelitian ini bertujuan menginventarisasi nematoda parasit dan identifikasi molekuler Meloidogyne pada tanaman padi lahan pasang surut di Kabupaten Banyuasin, Provinsi Sumatra Selatan. Sampel tanah dan akar dikumpulkan menggunakan metode purposive. Ekstraksi nematoda dari sampel tanah menggunakan metode flotasi sentrifugasi dan sampel akar menggunakan mist chamber. Identifikasi nematoda dilakukan berdasarkan karakter morfologi dan molekuler. Analisis komunitas nematoda dilakukan dengan menghitung nilai populasi absolut, frekuensi absolut, dan nilai prominensi. Identifikasi karakter molekuler dilakukan dengan primer D2A-D3B yang mengamplifikasi gen 28S rRNA dan analisis sekuensing DNA. Genus nematoda yang teridentifikasi yaitu Meloidogyne, Hirschmanniella, Tylenchus, dan Tylenchorhynchus. Meloidogyne merupakan genus yang mendominasi pada sampel akar dan tanah. Karakter molekuler Meloidogyne teramplifikasi dan menghasilkan pita berukuran ±750 pb. Analisis sekuensing menunjukkan kemiripan dengan Meloidogyne graminicola yang berkisar antara 98,90% hingga 99,45% dengan isolat dari beberapa negara di Genbank.
Rice (Oryza sativa L.) is one of the main staple food commodities with a strategic role in supporting national food security. One of the major constraints in rice cultivation is infestation of plant-parasitic nematodes, which can reduce plant growth and yield. This study aimed to inventory parasitic nematodes and molecular identification Meloidogyne on rice grown tidal swamp fields in Banyuasin Regency, South Province. Soil and root samples collected using purposively method. Nematodes were extracted from soil samples using the centrifugal flotation method, and from root samples using a mist chamber. Nematode identification was conducted based on morphological and molecular characteristics. Community analysis of nematodes was carried out by calculating absolute population, absolute frequency, and prominence value. Molecular identification was performed using D2A-D3B primers that amplify the 28S rRNA gene and DNA sequencing analysis. The identified nematode genera were Meloidogyne, Hirschmanniella, Tylenchus, and Tylenchorhynchus. Meloidogyne was the dominant genus in both root and soil samples. Molecular characterization of Meloidogyne produced an amplified band of approximately ±750 bp. DNA sequencing analysis revealed similarity with Meloidogyne graminicola, ranging from 98,90% to 99,45% compared with isolates from several countries deposited in GenBank.
