| Abstract: | Kanker merupakan permasalahan kesehatan global dengan prevalensi kematian mencapai 9,7 juta jiwa secara global pada tahun 2022. Kanker serviks merupakan jenis kanker dengan angka kejadian dan mortalitas tertinggi kedua di Indonesia setelah kanker payudara. Terapi konvensional memiliki keterbatasan berupa efek samping dan biaya tinggi, sehingga diperlukan alternatif yang lebih aman dan efektif. Spons laut (Porifera) mengandung senyawa bioaktif yang berpotensi sebagai antikanker, namun keanekaragaman spons di Nusa Penida dan potensi antikankernya belum banyak dieksplorasi. Penelitian ini bertujuan menentukan aktivitas antikanker beberapa sampel spons laut dari perairan Nusa Penida. Tahapan penelitian meliputi analisis spikula, ekstraksi, partisi, kromatografi kolom terbuka dan lapis tipis. Uji aktivitas antikanker dilakukan secara in vitro terhadap sel MCF-7 dan HeLa menggunakan uji microtetrazolium (MTT). Hasil menunjukkan bahwa fraksi etil asetat kode sampel NUP-008 memiliki profil senyawa potensial serta aktivitas sitotoksik tinggi, dengan nilai IC50 sebesar 122,67 µg/mL (MCF-7) dan 12,72 µg/mL (HeLa). Hasil ini menunjukkan potensi antikanker tinggi dan sumber antikanker yang menjanjikan. Cancer is a global health problem, with a global mortality rate of 9.7 million by 2022. Cervical cancer is the cancer with the second highest incidence and mortality rate in Indonesia after breast cancer. Conventional therapies have limitations in the form of side effects and high costs, so safer and more effective alternatives are needed. Marine sponges (Porifera) contain bioactive compounds with potential anticancer properties, but the diversity of sponges in Nusa Penida and their anticancer potential have not been widely explored. This study aims to determine the anticancer activity of several marine sponge samples from Nusa Penida waters. The research stages include spicule analysis, extraction, partitioning, open column and thin layer chromatography. Anticancer activity tests were carried out in vitro on MCF-7 and HeLa cells using the MTT assay. The results showed that the ethyl acetate fraction of NUP-008 sample code had a potential compound profile and high cytotoxic activity, with IC50 values of 122.67 µg/mL (MCF-7) and 12.72 µg/mL (HeLa). These results indicate moderate to high anticancer potential and the potential of Nusa Penida marine sponges as a promising anticancer source. |
