| Abstract: | Penelitian ini bertujuan mengevaluasi konsentrasi optimal BAP (Benzyl amino purine) untuk mendukung pertumbuhan Desmanthus virgatus pada kultur in vitro melalui kultur jaringan. Penelitian dirancang menggunakan Rancangan Acak Lengkap (RAL) yang terdiri atas 5 perlakuan dan masing-masing perlakuan memiliki 20 ulangan. Perlakuan yang digunakan adalah P0 (kontrol), P1 (BAP 0,5 ppm), P2 (BAP 1 ppm), P3 (BAP 1,5 ppm), P4 (BAP 2 ppm). Peubah yang diamati meliputi tinggi tanaman, jumlah daun, jumlah cabang, jumlah ranting, jumlah kerontokan daun, dan warna daun. Data dianalisis menggunakan Analysis of Variance (ANOVA) dan diuji lanjut dengan uji Duncan. Hasil penelitian menunjukkan bahwa tanaman D. virgatus yang ditambahkan BAP 1,5 ppm berpengaruh nyata (P<0,05) terhadap tinggi tanaman, jumlah cabang dan jumlah daun yang dihasilkan dibandingkan kontrol, serta menghasilkan visual warna hijau daun yang normal (7,GY 6/8 dan 5GY 7/8). Penambahan BAP pada konsentrasi berbeda tidak berpengaruh nyata (P>0,05) terhadap jumlah ranting yang dihasilkan. Penambahan BAP dengan konsentrasi 1,5 ppm menjadi dosis yang paling optimal untuk pertumbuhan Desmanthus virgatus melalui kultur jaringan. This study aimed to evaluate the optimal concentration of BAP (Benzyl amino purine) to support the growth of Desmanthus virgatus in vitro through tissue culture. The research was designed using a Completely Randomized Design (CRD), consisting of 5 treatments with 20 replications for each treatment. The treatments used were P0 (control), P1 (BAP 0.5 ppm), P2 (BAP 1 ppm), P3 (BAP 1.5 ppm), and P4 (BAP 2 ppm). The observed variables included plant height, number of leaves, number of branches, number of shoots, leaf drop, and leaf color. Data were analyzed using Analysis of Variance (ANOVA), and further tested with Duncan's test. The results showed that D. virgatus plants added with 1.5 ppm BAP had a significant effect (P<0.05) on plant height, number of branches and number of leaves produced compared to the control, and produced a normal green leaf color (7.5GY 6/8 and 5GY 7/8). The addition of BAP at different concentrations had no significant effect (P>0.05) on the number of branches produced. The addition of BAP with a concentration of 1.5 ppm was the most optimal dose for the growth of Desmanthus virgatus through tissue culture. |
