Abstract: Kentang kultivar IPB CP3 telah dikembangkan di Pusat Bioteknologi IPB sebagai kultivar lokal untuk industri keripik maupun french fries. Namun, kultivar tersebut masih perlu diperbaiki tingkat toleransinya terhadap penyakit. Upaya perbaikan kultivar IPB CP3 telah dilakukan melalui mutasi, tetapi perlu dilanjutkan dengan analisis molekuler untuk memastikan ada perubahan genetik. Penelitian ini bertujuan menganalisis mutan kentang IPB CP3 yang telah diirradiasi oleh sinar gamma menggunakan marka Random Amplified Polymorphic DNA (RAPD) dan Inter Simple Sequence Repeat (ISSR). Amplifikasi DNA tanaman kentang dilakukan menggunakan primer RAPD dan ISSR. Hasil amplifikasi menunjukkan polimorfisme sebesar 44%. Hal ini menunjukkan radiasi sinar gamma menghasilkan variasi genetik. Sebanyak 11 mutan yang dianalisis, mutan klon 20.29 memiliki hubungan terjauh berdasarkan dendogram UPGMA. Primer OPA 2 memberikan kontribusi paling baik dalam mendeteksi perbedaan yang terlihat pada scatter plot PCA.
The potato cultivar IPB CP3 has been developed at the IPB Biotechnology Center as a local cultivar for the chip and French fries industries. However, this cultivar still requires improvement in its tolerance to diseases. Improvement efforts for the IPB CP3 cultivar have been carried out through mutation, but further molecular analysis is necessary to confirm genetic changes. This study aimed to analyze IPB CP3 potato mutants irradiated with gamma rays using Random Amplified Polymorphic DNA (RAPD) and Inter Simple Sequence Repeat (ISSR) markers. DNA amplification of potato plants was performed using RAPD and ISSR primers. The amplification results showed a polymorphism rate of 44%. This indicated that gamma-ray irradiation generated genetic variation. Among the 11 mutants analyzed, mutant clone 20.29 exhibited the most distant relationship based on the UPGMA dendrogram. The OPA 2 primer contributed most effectively in detecting differences as shown in the PCA scatter plot.
