| Abstract: | Kumis kucing (Orthosiphon aristatus) banyak digunakan sebagai bahan baku
obat tradisional dan pemanfaatannya dipengaruhi oleh variasi kandungan kimianya.
Penelitian ini dilakukan untuk mengkarakterisasi profil kimia dan aktivitas
antibakteri daun kumis kucing yang diperoleh dari dua lokasi (Bogor dan
Sukabumi). Sampel dianalisis menggunakan DART-TOFMS, aktivitas antibakteri
ekstrak terhadap Staphylococcus aureus dan Escherichia coli, dan analisis
kemometrik (PCA dan OPLS-DA). Sebanyak 18 senyawa kimia terdeteksi,
termasuk 2 senyawa unik yang hanya terdapat pada sampel Bogor dan 5 senyawa
unik yang hanya terdapat pada sampel Sukabumi. Ekstrak dari Sukabumi
menghasilkan zona hambat yang lebih besar dibandingkan dengan ekstrak dari
Bogor. Analisis PCA dan OPLS-DA mengungkapkan pemisahan spektrum kimia
yang jelas antara kedua lokasi. DART-TOFMS yang dikombinasikan dengan
kemometrik terbukti efektif dan akurat untuk karakterisasi cepat sebagai pendukung
pengendalian kualitas kumis kucing sebagai bahan baku obat herbal. Kumis kucing (Orthosiphon aristatus) was widely used as a raw material for traditional medicine and its utilization was influenced by the variation in chemical constituents. This study was conducted to characterize the chemical profiles and antibacterial activities of kumis kucing leaves obtained from two locations (Bogor and Sukabumi). The samples were analyzed using DART-TOFMS, antibacterial activity of the extracts against Staphylococcus aureus and Escherichia coli was assessed, and chemometric analyses (PCA and OPLS-DA) were performed. A total of 18 compounds were detected, including 2 compounds unique to Bogor samples and 5 compounds unique to Sukabumi samples. Extracts from Sukabumi produced larger inhibition zones compared to those from Bogor. PCA and OPLS DA analyses revealed a clear separation of chemical spectra between the two locations. It was demonstrated that DART-TOFMS combined with chemometrics was an effective and accurate approach for rapid characterization, supporting the quality control of kumis kucing as a raw material for herbal medicine. |
