Abstract: Penelitian ini bertujuan mengevaluasi suplemen pakan diperkaya asam lemak tak jenuh ganda (PUFA) dan antioksidan alami dalam bentuk enkapsulasi melalui pendekatan fermentabilitas rumen in vitro dan profil metabolomik. Tiga perlakuan suplementasi diuji secara in vitro, yaitu: PUFA nabati dan antioksidan alami (R1), sabun kalsium PUFA nabati dan antioksidan alami (R2), serta enkapsulasi PUFA nabati dan antioksidan alami (R3), dengan rancangan acak kelompok (RAK). Hasil menunjukkan bahwa semua bentuk suplementasi tidak memberikan perbedaan yang nyata (P>0,05) terhadap pH rumen, populasi bakteri dan protozoa, konsentrasi Volatile Fatty Acid total, serta konsentrasi amonia antar perlakuan. Namun demikian, perlakuan R3 secara signifikan menurunkan (P<0,05) persentase asam iso-valerat. Enkapsulasi juga menurunkan (P<0,05) degradasi bahan kering dan bahan organik dalam rumen serta menurunkan kecernaan bahan kering pasca rumen dibandingkan perlakuan lainnya. Senyawa metabolit yang teridentifikasi pada perlakuan enkapsulasi meliputi talatizamin serta dipeptida leucine-leucine dan leucine-phenylalanine, yang merupakan komponen yang berkontribusi dalam formulasi bahan enkapsulan. Analisis metabolomik mengonfirmasi bahwa proses enkapsulasi mampu melindungi senyawa bioaktif baik di dalam rumen maupun pasca-rumen. Dengan demikian, suplementasi antioksidan alami yang diperkaya PUFA dalam bentuk enkapsulasi secara efektif melindungi PUFA dan senyawa antioksidan dari degradasi oleh mikroba rumen, serta berpotensi digunakan sebagai suplemen berenergi tinggi untuk mendukung fase reproduksi pada ruminansia.
The present study aimed to evaluate the encapsulated PUFAs-enriched natural antioxidant using in vitro rumen fermentability and metabolomic profile approaches. Three supplementation treatments were tested in vitro: Non encapsulated PUFAs and natural antioxidant (R1), calcium soap of PUFAs and natural antioxidant (R2), and encapsulated PUFAs-enriched natural antioxidant (R3) using a completely randomized block design. The results showed that all supplementation forms did not show significant differences (P>0.05) in rumen pH, bacterial and protozoa populations, total Volatile Fatty Acid concentration, and ammonia concentration among treatments. However, R3 significantly decreased (P<0.05) the iso-valeric acid percentage. The encapsulation form reduced (P<0.05) ruminal dry matter and organic matter degradability, and post-ruminal dry matter digestibility compared to the others. The metabolites identified in the encapsulation treatment included talatizamine, as well as leucine-leucine and leucine phenylalanine, which are components involved in the formulation of the encapsulating material. The metabolomic analysis confirmed that encapsulation preserved bioactive compounds in the rumen and post-ruminal. In conclusion, the supplementation of encapsulated PUFAs-enriched natural antioxidant effectively protects PUFAs and natural antioxidant compounds from rumen microbial degradation, supporting their potential use as a high-energy supplement to support the reproductive phase in ruminants.
