| Abstract: | Anggrek Thrixspermum malayanum J.J. Smith. merupakan anggrek yang
memiliki persebaran di pulau Jawa namun masih belum banyak diteliti dalam
perbanyakannya melalui biji dalam kultur in vitro. Perkecambahan dan
pertumbuhan benih anggrek dalam kultur in vitro sangat dipengaruhi jenis media
dasar, sedangkan bahan organik kompleks dapat digunakan sebagai suplemen
pertumbuhan tanaman in vitro. Penelitian ini bertujuan untuk mendapatkan
kombinasi media basal dan bahan organik kompleks terbaik untuk pertumbuhan
anggrek T. malayanum secara in vitro. Penelitian dilakukan di Laboratorium
Fakultas Pertanian IPB University. Protocorm yang berasal dari semaian biji
digunakan sebagai bahan tanam dan diujikan ke 4 taraf media dasar yaitu ½ MS,
KC, Growmore, dan Gaviota, yang dikombinasikan dengan 3 taraf bahan organik
kompleks yaitu pisang, tomat, dan pepton. Percobaan dilaksanakan menggunakan
rancangan kelompok lengkap teracak dengan 3 ulangan. Setiap ulangan terdiri dari
3 cawan petri dan setiap cawan berisi 12 protocorm. Pertumbuhan protocorm
diamati selama 22 minggu setelah kultur (MSK) dengan peubah yang diamati yaitu
persentase protocorm hidup, membentuk tunas, membentuk rambut akar,
membentuk daun, membentuk protocorm sekunder, bertunas sekunder, dan ratarata
jumlah daun per-protocorm. Persentase protocorm hidup tertinggi teramati
pada media ½ MS dan KC dengan tambahan pisang dan tomat 150 g l-1. Jumlah
protocorm yang berhasil membentuk daun dan akar teramati pada media dasar ½
MS dan KC dengan persentase pada masing-masing media sebesar 17,82% dan
5,84% membentuk daun, dan sebesar 8,20% dan 4,36% membentuk akar. Hasil
penelitian ini menghasilkan protokol yang optimum untuk regenerasi protocorm T.
malayanum J.J. Sm. menjadi planlet. Thrixspermum malayanum J.J.Smith. orchid habited area in Java island and currently remains underreppresented in in vitro propagation by seed.. Germination and growth of orchid in vitro is highly infuluenced by basal medium, while organic compound additives could be used to supplement in vitro plant growth. This research aimed to formulate optimum combination of basal medium and complex organic additives for T. malayanum orchid in vitro growth. This research was conducted in laboratory Faculty of Agriculture, IPB University. Seed sown protocorm was used as plant material in this research which then tested in treatment medium containing combination of 4 basal medium in which are ½ MS, KC, Growmore, and Gaviota, and 3 organic complex material which are banana, tomato, and pepton. The experiment used randomized complete block design with 3 replications. Each replication consists of 3 petri dishes with 12 protocorms per dish. The protocorm were observed for 22 weeks after culture (WAC) for their growth parameter including percentage of alive protocorm; bud forming; root hair forming; leaf forming; secondary protocorm forming; secondary bud forming; and mean leaf count per protocorm. Highest percentage of alive plant obtained using ½ MS and KC basal medium supplemented with banana and tomato 150 g l-1. The ½ MS and KC medium has the most successful rate in growing protocorm into fully formed plantlet with leaf formed percentage was 17,82% and 5,84%; and root formed percentage was 8,2% and 4,36%. These results give an insight for optimum protocol for protocorm regeneration of T. malayanum J.J. Sm. into fully developed plantlet. |
