Abstract: Metabolit sekunder yang dihasilkan oleh bakteri laut berasosiasi spons memiliki potensi besar sebagai sumber senyawa bioaktif, seperti antibakteri dan antikanker. Penelitian ini bertujuan untuk mengevaluasi aktivitas bioaktif dari Kocuria marina galur SAB E28 melalui uji antibakteri, uji sitotoksisitas, serta identifikasi senyawa kimia penyusun ekstraknya. Tahapan penelitian meliputi karakterisasi morfologi bakteri dan dilanjutkan ekstraksi senyawa bioaktif dengan etil asetat. Ekstrak kasar etil asetat bakteri digunakan dalam pengujian aktivitas antibakteri melalui metode difusi sumuran terhadap beberapa bakteri uji dan toksisitas dengan Brine Shrimp Lethality Test (BSLT). Senyawa kimiawi penyusun ekstrak kasar etil asetat dianalisis menggunakan Gas Chromatography-Mass Spectrometry (GC-MS). Karakterisasi morfologi menunjukkan bahwa koloni bakteri berpigmen kuning dan tergolong bakteri Gram positif dengan bentuk sel diplokokus. Ekstrak kasar etil asetat menunjukkan daya hambat kategori sedang terhadap bakteri Escherichia coli dan Pseudomonas aeruginosa dengan diameter zona hambat berturut-turut 8,85 mm dan 8,70 mm. Uji tokisisitas menghasilkan nilai LC50 sebesar 92,11 µg/mL dengan rata-rata persentase kematian larva sebesar 53,57%. Analisis senyawa kimia penyusun ekstrak dengan GC-MS di antaranya teridentifikasi sebagai Benzeneacetic acid, Phenyllactic acid, dan n-Hexadecanoic acid yang berpotensi memiliki bioaktivitas sebagai antibakteri dan antikanker. Temuan ini mengindikasikan bahwa K. marina galur SAB E28 merupakan kandidat menjanjikan dalam pencarian sumber senyawa antibakteri dan antikanker baru.
Sponge-associated marine bacteria are recognized for their ability to produce secondary metabolites with promising biological activities, including antibacterial and anticancer effects. This study investigates the bioactive potential of Kocuria marina strain SAB E28 through antibacterial and cytotoxicity assessments, as well as chemical profiling of its crude extract. The research commenced with morphological characterization and followed by metabolite extraction using ethyl acetate. Antibacterial activity was assessed via well diffusion assays against several test bacteria, while toxicity was evaluated using the brine shrimp lethality test. Chemical constituents were analyzed through gas chromatography–mass spectrometry (GC-MS). Morphological characterization revealed yellow pigmented colonies identified as Gram-positive diplococci. The ethyl acetate crude extract exhibited moderate inhibition activity, with inhibition zones of 8,85 mm and 8,70 mm against Escherichia coli and Pseudomonas aeruginosa, respectively. Toxicity analysis yielded an LC50 value of 92,11 µg/mL and a mean larval mortality of 53,57%. GC-MS analysis identified several compounds including benzeneacetic acid, phenyllactic acid, and n-hexadecanoic acid, which are recognized for their bioactive potential. These findings suggest that K. marina strain SAB E28 is a promising candidate for the discovery of novel antibacterial and anticancer agents.
