Abstract: Pengembangan antibodi rekombinan scFv-Fc anti-HER2 merupakan alternatif terapeutik untuk kanker payudara HER2+. Format antibodi ini memiliki keunggulan yaitu seluruh domain penyusunnya tersusun dalam satu struktur gen yang ringkas, sehingga memungkinkan ekspresi gen yang efisien dalam sel inang non-mamalia seperti yeast. Penelitian ini bertujuan memperbanyak plasmid ekspresi penyandi gen antibodi scFv-Fc anti-HER2 dalam Escherichia coli dan mentransformasikannya ke yeast Komagataella phaffii sebagai sistem ekspresi eukariotik. Plasmid pJ912-anti-HER2 berhasil diperbanyak melalui transformasi pada E. coli DH5a dengan metode kejut panas dan diperoleh konsentrasi cukup tinggi dengan kemurnian yang baik pada rasio A260/280 dan A260/230. Transformasi pJ912-anti-HER2 ke K. phaffii melalui metode kejut listrik menghasilkan koloni transforman putatif dengan efisiensi transformasi yang masih rendah. Keberhasilan integrasi plasmid menunjukkan potensi sistem ini untuk produksi antibodi rekombinan secara efisien dan ekonomis.
The development of recombinant scFv-Fc anti-HER2 antibodies is a therapeutic alternative for HER2+ breast cancer. This antibody format has the advantage that all the constituent domains are arranged in one compact gene structure, allowing efficient gene expression in non-mammalian host cells such as yeast. This study aims to propagate the scFv-Fc anti-HER2 gene expression plasmid in Escherichia coli and transform it into Komagataella phaffii yeast as a eukaryotic expression system. Plasmid pJ912-anti-HER2 was successfully amplified through transformation in E. coli DH5a using the heat shock method and obtained a sufficiently high concentration with good purity at the A260/280 and A260/230 ratios. Transformation of pJ912-anti-HER2 into K. phaffii using the electroporation method produced putative transformant colonies with low transformation efficiency. The successful integration of plasmids shows the potential of this system for efficient and economical production of recombinant antibodies.
