| Abstract: | Perairan Banggai merupakan salah satu perairan yang menjadi lokasi
upwelling. Terjadinya upwelling membuat banyak nutrien yang berada di perairan
dalam naik ke permukaan. Penelitian ini bertujuan untuk mengisolasi dan
mengkarakterisasi dan menghitung kelimpahan bakteri di Perairan Banggai. Isolasi
bakteri dilakukan untuk melihat kelimpahan bakteri menggunakan pengenceran
bertingkat dengan media padat Zobell 2216E. Kelimpahan bakteri di inti upwelling
dan non-upwelling menunjukkan hasil yang berbeda, yaitu kelimpahan tinggi
berada di perairan inti upwelling. Hasil isolasi bakteri diperoleh 26 isolat.
Identifikasi 26 isolat bakteri dilakukan melalui pewarnaan Gram dan uji fisiologis
bakteri terisolasi, yaitu uji katalase, uji motilitas, uji oksidase, uji sitrat, uji hidrolisis
pati, uji TSIA, uji indol, uji lisin, dan uji methyl red. Hasil identifikasi terhadap 26
isolat menunjukkan kecocokan dengan genus Acinetobacter, Alcaligenes,
Alteromonas, Brucella, Enterococcus, Halomonas, Marinococcus, Morococcus,
Pseudomonas, dan Staphylococcus. Banggai waters are one of the waters that is a location for upwelling. The occurrence of upwelling causes many nutrients in deep waters to rise to the surface. This research aims to isolate, characterize and quantify the abundance of bacteria in Banggai waters. Bacterial isolation was carried out to see the abundance of bacteria using multilevel dilution with Zobell 2216E solid media. The abundance of bacteria in upwelling and non-upwelling cores shows different results, namely high abundance in upwelling core waters. The results of bacterial isolation obtained 26 isolates. Identification of 26 bacterial isolates was carried out through Gram staining and physiological tests of isolated bacteria, namely catalase test, motility test, oxidase test, citrate test, starch hydrolysis test, TSIA test, indole test, lysine test, and methyl red test. The identification results of 26 isolates showed matches with the genera Acinetobacter, Alcaligenes, Alteromonas, Brucella, Enterococcus, Halomonas, Marinococcus, Morococcus, Pseudomonas, and Staphylococcus. |
