| dc.description.abstract | Peptida antimikroba (AMP) merupakan senyawa alami yang berpotensi sebagai alternatif antibiotik untuk mengatasi resistensi bakteri. Salah satu AMP yang menjanjikan adalah snakin, yang dikenal memiliki aktivitas luas dan kandungan sistein tinggi. Penelitian ini bertujuan mengisolasi DNA dan mengidentifikasi gen penyandi snakin pada tanaman famili Solanaceae yang tumbuh di Indonesia, seperti leunca, kentang, cabai merah, cabai rawit, dan terung bulat. Isolasi DNA dilakukan menggunakan metode CTAB dengan perlakuan nitrogen cair, RNAse, dan Proteinase K. Analisis menggunakan spektrofotometer Nanodrop (A260/A230 dan A260/A280) dan elektroforesis menunjukkan bahwa perlakuan tersebut menghasilkan kemurnian dan konsentrasi DNA yang ideal umtuk dilakukam PCR. Amplifikasi PCR menggunakan primer SN1 dilakukan dengan variasi suhu annealing untuk optimasi, dan hasilnya divisualisasikan melalui elektroforesis gel agarosa. Suhu annealing optimum ditemukan pada 53 °C dengan amplikon spesifik berukuran 680 bp. Gen snakin berhasil teridentifikasi pada cabai merah dan leunca, tetapi tidak terdeteksi pada kentang, cabai rawit dan terung bulat. Hasil ini menunjukkan bahwa keberadaan gen snakin bervariasi antar spesies Solanaceae. Penelitian ini mendukung potensi pengembangan AMP dari tanaman lokal sebagai agen antibakteri alternatif. | - |
| dc.description.abstract | Antimicrobial peptides (AMPs) are natural compounds with promising potential as antibiotic alternatives to combat bacterial resistance. One notable AMP is snakin, known for its broad-spectrum activity and high cysteine content. This study aimed to isolate DNA and identify the snakin-encoding gene in Solanaceae plants commonly found in Indonesia, including black nightshade (leunca), potato, red chili, bird’s eye chili, and round eggplant. DNA isolation was carried out using the CTAB method, with additional treatment using liquid nitrogen, RNase, and Proteinase K. Analysis using a Nanodrop spectrophotometer (A260/A230 and A260/A280) and gel electrophoresis showed that these treatments produced DNA with suitable purity and concentration for PCR. PCR amplification was performed using SN1 primers, with various annealing temperatures to determine optimal conditions, and the results were visualized using agarose gel electrophoresis. The optimal annealing temperature was found to be 53?°C, producing a specific amplicon of 680 bp. The snakin gene was successfully identified in red chili and black nightshade, but not detected in potato, bird’s eye chili, or round eggplant. These findings suggest that the presence of the snakin gene varies among Solanaceae species. This study supports the potential development of AMPs from local plants as alternative antibacterial agents. | - |
