dc.description.abstractBudidaya bawang merah memiliki permasalahan utama yaitu penyakit layu fusarium yang disebabkan oleh cendawan Fusarium oxysporum. Pengendalian penyakit layu fusarium dapat menggunakan fungisida dan teknik pengendalian biologis menggunakan bakteri endofit. Penelitian ini bertujuan mengetahui strain bakteri endofit yang dapat meningkatkan ketahanan bawang merah terhadap serangan penyakit layu fusarium yang disebabkan oleh cendawan F. oxysporum di Rumah Kaca dan mampu memproduksi umbi setelah pengujian. Rancangan percobaan yang digunakan dalam penelitian ini adalah Rancangan Acak Lengkap (RAL) faktor tunggal dengan 5 taraf perlakuan, yaitu umbi benih bawang merah varietas Bima (asal penangkar) yang ditanam di media tanpa infeksi buatan (K*) dan empat taraf perlakuan ditanam di media infeksi buatan cendawan F. oxysporum, yaitu umbi benih bawang merah varietas Bima (asal penangkar) (K), umbi G1 varietas Bima tanpa diperkaya bakteri endofit (asal kultur jaringan) (Ki), umbi G1 varietas Bima diperkaya bakteri endofit strain 35 (B35), dan umbi G1 varietas Bima diperkaya bakteri endofit 39 (B39). Uji ketahanan dilakukan dengan penambahan isolat F. oxysporum dalam kultur sorgum pada media tanam. Pengamatan dilakukan terhadap ketahanan, jumlah helai daun, tunas, akar, dan bobot umbi. Hasil penelitian menunjukkan bakteri strain 39 meningkatkan ketahanan terhadap layu fusarium sebesar 82% dan bakteri strain 35 meningkatkan sebesar 40% dibandingkan umbi G1 tanpa pengkayaan bakteri endofit. Secara umum bakteri endofit dapat meningkatkan pertumbuhan bawang merah seperti jumlah helai daun, jumlah tunas, dan bobot umbi. Bobot umbi bakteri endofit strain 35 dan 39 memiliki peningkatan bobot sebesar 61% dan 134% dari umbi tanpa bakteri endofit yang hanya memiliki peningkatan bobot sebesar 49%.-
dc.description.abstractShallot cultivation has a major problem, namely fusarium wilt disease caused by the fungus Fusarium oxysporum. Control of Fusarium wilt can be achieved using fungicides and biological control techniques utilizing endophytic bacteria. This research aims to identify endophytic bacterial strains that can increase shallot resistance to F. oxysporum attack in the greenhouse and enable tuber production after testing. The experimental design used in this study was a single-factor Completely Randomized Design (CRD) with 5 treatment levels. Bima variety shallot seeds (sourced from a breeder) planted in soil media (K*) and four treatment levels planted artificial F. oxysporum infection media, namely Bima variety shallot seeds (sourced from a breeder) (K), Bima G1 Bima variety tubers without endophytic bacterial enrichment (from tissue culture) (Ki), Bima variety G1 bulbs enriched with endophytic bacteria strain 35 (B35), and Bima variety G1 bulbs enriched with endophytic bacteria 39 (B39). Resistance was tested by adding F. oxysporum isolate in sorghum culture to the planting medium. Observations included resistance levels, number of leaves, shoots, roots, and tuber weight. The results showed that bacterial strain 39 increased resistance to Fusarium wilt by 82%, while bacterial strain 35 increased it by 40% compared to G1 tubers without endophytic bacterial enrichment. Overall, endophytic bacteria were found to enhance shallot growth, specifically increasing the number of leaves, shoots, and tuber weight. Regarding tuber weight, endophytic bacterial strains 35 and 39 showed an increase of 61% and 134%, respectively, compared to tubers without endophytic bacteria was only 49%.-
