Abstract: Bunga rosella merah (Hibiscus sabdariffa L.) merupakan tanaman yang memiliki banyak khasiat dan perlu diteliti tingkat keamanan penggunaannya. Penelitian ini bertujuan untuk menganalisis tingkat toksisitas akut infusa bunga rosella merah (IBRM) pada mencit betina dengan metode penentuan nilai lethal dose 50 (LD50) dan mengetahui dampak pemberian IBRM terhadap fisiologis tubuh. Infusa bunga rosella merah diberikan dengan dosis 0, 1, 5, 10, dan 15 g/kgBB secara peroral. Parameter yang diamati, yaitu gejala klinis, respons fisiologis, bobot badan, bobot organ absolut, bobot organ relatif, dan mortalitas mencit selama 14 hari. Pengamatan setelah diberikan IBRM didapatkan hasil bahwa mencit tidak mengalami kematian, tidak menimbulkan kelainan gejala klinis, dan tidak terjadi perubahan pada organ secara makroskopis. Uji secara statistik menunjukkan hasil tidak berbeda nyata (p>0,05) pada parameter bobot badan, suhu tubuh, frekuensi detak jantung, dan bobot organ absolut. Hasil berbeda nyata (p<0,05) didapatkan pada pengujian terhadap parameter frekuensi napas, bobot organ relatif paru-paru, dan bobot organ relatif hati. Berdasarkan penelitian didapatkan nilai LD50 dari IBRM, yaitu >15 g/kgBB dan termasuk ke dalam kategori praktis tidak toksik
The red rosella flower (Hibiscus sabdariffa L.) is a plant with many properties that needs to be studied to determine its safety level. This study aims to analyze the acute toxicity level of red rosella flower infusion in female mice by determining the lethal dose 50 (LD50) value and assessing its impact on physiological body functions. The red rosella infusion was administered orally at doses of 0, 1, 5, 10, and 15 g/kgBB. The following parameters were observed clinical symptoms, physiological responses, body weight, absolute organ weight, relative organ weight, and mortality of the mice over the course of 14 days. After receiving red rosella flower infusion, the mice did not die, exhibit clinical symptoms, or show macroscopic organ changes. Statistical tests showed that the results were not significantly different (p>0,05) for body weight, body temperature, heart rate, and absolute organ weight. Significant differences (p<0,05) were found in the breathing frequency, relative lung weight, and relative liver weight parameters. Based on this research, the LD50 value of red rosella flower infusion was determined to be greater than 15 g/kgBB, placing it in the practically non-toxic category.
