| Abstract: | Kebutuhan domba meningkat seiring dengan bertambahnya jumlah penduduk dan semakin meningkatnya permintaan akan protein hewani. Upaya peningkatan kebutuhan domba dapat dilakukan dengan memanfaatkan teknologi reproduksi, seperti inseminasi buatan (IB). Penelitian ini bertujuan untuk menganalisis potensi dan efektivitias penggunaan KOSR sebagai nutrisi semen cair domba terhadap motilitas dan keutuhan membran plasma spermatozoa domba Garut. Media pengencer dibuat dengan 3 perlakuan, yaitu without KOSR (P1), 5% (P2) dan 10% (P3). Perlakuan diamati dengan waktu pengamatan 0 jam, 24 jam, 48 jam, 72 jam dan 96 jam. Data dianalisis statistik dengan ANOVA satu arah dan dilanjutkan dengan uji Duncan. Interaksi antara perlakuan dan lama penyimpanan memiliki pengaruh yang sangat signifikan terhadap motilitas dan keutuhan membran plasma spermatozoa. P3 dengan 10% KOSR memberikan hasil terbaik di setiap jam pengamatan. Waktu penyimpanan paling optimal dari penelitian ini adalah 72 jam. Hasil penelitian menunjukkan penggunaan KOSR dalam pengencer semen domba Garut berdampak positif terhadap motilitas dan keutuhan membran plasma. The need for sheep increases along with the increasing population and the increasing demand for animal protein. Efforts to increase the need for sheep can be done by utilizing reproductive technology, such as artificial insemination (AI). This study aims to analyze the potential and effectiveness of the use of KOSR as a liquid sheep semen nutrient on the motility and integrity of the plasma membrane of Garut sheep spermatozoa. The diluent media was made with 3 treatments, namely without KOSR (P1), 5% (P2) and 10% (P3). The treatments were observed with observation times of 0 hours, 24 hours, 48 hours, 72 hours and 96 hours. Data were statistically analyzed using one-way ANOVA and continued with the Duncan test. The interaction between treatment and storage time had a very significant effect on the motility and integrity of the plasma membrane of spermatozoa. P3 with 10% KOSR gave the best results in every hour of observation. The most optimal storage time of this study was 72 hours. The results showed that the use of KOSR in Garut sheep semen diluents had a positive impact on the motility and integrity of the plasma membrane. |
