Abstract: Pengenceran semen membutuhkan bahan pengencer yang tepat dalam mempertahankan kualitas spermatozoa. Penelitian dilakukan untuk mengevaluasi efektivitas penggunaan serum domba dan l-glutamine pada pengencer semen cair domba dalam mempertahankan motilitas dan keutuhan membran plasma. Penelitian dilakukan dengan penambahan nutrisi pada bahan pengencer buffer Tris, soya lesitin, dan l-glutamine dengan berbagai konsentrasi yaitu serum domba 5% dan 10% serta kontrol yaitu tanpa penambahan serum domba (0%). Parameter motilitas diamati selama 4 hari setiap 24 jam, yaitu 0 jam, 24 jam, 48 jam, 72 jam, dan 96 jam. Parameter keutuhan membran plasma dilakukan pada kurun waktu 96 jam. Pengujian statistik dengan menggunakan metode one-way analysis of variance (ANOVA) plus Duncan pada aplikasi SPSS menunjukkan bahwa penambahan serum domba dan l-glutamine secara nyata (P<0,05) dapat mempertahankan motilitas spermatozoa namun berbanding terbalik terhadap keutuhan membran plasma (P>0,05). Uji lanjutan menunjukkan bahwa penambahan serum domba dan l-glutamine pada perlakuan P3 dengan konsentrasi 10% memberikan hasil yang lebih baik terhadap motilitas dan keutuhan membran plasma.
Semen dilution requires an appropriate diluent to preserve the quality of spermatozoa. This study aimed to evaluate the effectiveness of sheep serum and L-glutamine in a liquid semen diluent for Garut sheep in maintaining sperm motility and plasma membrane integrity. The diluent used was based on Tris buffer supplemented with soy lecithin and L-glutamine, with sheep serum added at concentrations of 0% (control), 5%, and 10%. Sperm motility was assessed over a 4-day storage period at 24-hour intervals (0, 24, 48, 72, and 96 hours), while plasma membrane integrity was evaluated at 96 hours. Statistical analysis using one-way ANOVA followed by Duncan’s test (SPSS) showed that the addition of sheep serum and L-glutamine significantly (P<0.05) maintained sperm motility, but had no significant effect (P>0.05) on plasma membrane integrity. Further analysis indicated that the treatment with 10% sheep serum (P3) provided the best results in preserving both motility and plasma membrane integrity.
