| Abstract: | Oosit yang kompeten merupakan salah satu faktor yang menentukan keberhasilan produksi embrio in vitro. Penelitian ini bertujuan untuk menganalisis efektifitas penambahan glutation pada sistem pematangan dan fertilisasi dalam meningkatkan kompetensi oosit domba secara in vitro. Penelitian ini terdiri dari dua tahap. Tahap pertama merupakan maturasi oosit in vitro. Pada tahap ini, oosit dibagi menjadi empat kelompok dan dimatangkan dalam medium maturasi yang masing-masing ditambahkan GSH 0 mM (kontrol), GSH 0,5 mM, GSH 1,0 mM, dan GSH 1,5 mM. Oosit domba dimatangkan selama 24 jam dalam inkubator CO2 5% dengan suhu 38,5°C. Tahap kedua yaitu fertilisasi in vitro. Tahap ini dibagi menjadi tiga kelompok antara lain penambahan GSH 1,0 mM (dosis optimum yang diperoleh pada penelitian tahap I) pada medium maturasi (IVM+; IVF-), medium fertilisasi (IVM-; IVF+), serta kombinasi medium maturasi dan fertilisasi (IVM+; IVF+). Oosit dan sperma kemudian di ko-inkubasi selama 14 jam dalam inkubator CO2 5% dengan suhu 38,5°C. Tingkat maturasi dan fertilisasi oosit dievaluasi menggunakan pewarnaan aceto-orcein 2%. Hasil penelitian tahap pertama menunjukkan bahwa penambahan GSH 1,0 mM pada medium maturasi (87,00%) menghasilkan persentase oosit mencapai MII secara signifikan lebih tinggi (P<0,05) dibandingkan dengan kelompok GSH 0,5 mM (73,41%), GSH 1,5 mM (78,23%), dan kontrol (70,50%). Hasil penelitian tahap kedua menunjukkan bahwa persentase tingkat fertilisasi normal (2PN) pada kelompok yang ditambahkan GSH dalam medium IVM (56,43%) secara signifikan lebih tinggi (P<0,05) dibandingkan kelompok yang ditambahkan GSH pada medium kobinasi maturasi dan fertilisasi (18,45%). Oleh karena itu, dapat disimpulkan bahwa penambahan glutation 1,0 mM pada media maturasi lebih efektif dalam meningkatkan kompetensi oosit domba untuk menjadi matang dan menghasilkan tingkat fertilisasi lebih baik. Oocyte competence is a primary factor for the success of embryo production in vitro. This study aimed to analyze the effectiveness of adding glutathione to the maturation and fertilization system in increasing the competence of sheep oocytes in vitro. This study consists of two stages. The first stage is in vitro maturation. At this stage, oocytes are divided into four groups and matured in a maturation medium, each added with 0 mM GSH (control), 0.5 mM GSH, 1.0 mM GSH, and 1.5 mM GSH. Sheep oocytes are matured for 24 hours in a 5% CO2 incubator at 38.5°C. The second stage is in vitro fertilization. This stage is divided into three groups, including the addition of 1.0 mM GSH (the optimum dose obtained in the first stage of the study) to the maturation medium (IVM+; IVF-), fertilization medium (IVM-; IVF+), and a combination of maturation and fertilization medium (IVM+; IVF+). The oocytes and sperm were co-incubated for 14 hours in a 5% CO2 incubator at 38.5°C. The oocyte maturation and fertilization rate were evaluated using 2% aceto-orcein staining. The results of the first stage of the study showed that the addition of 1,0 mM GSH in the maturation medium (87.00%) resulted in a significantly higher (P < 0.05) percentage of oocytes reaching MII compared to the 0.5 mM GSH (73.41%), 1.5 mM GSH (78.23%), and control (70.50%) groups. The results of the second stage of the study showed that the percentage of normal fertilization rate (2PN) in the group added with GSH in IVM medium (56.43%) was significantly higher (P < 0.05) than the group added with GSH in the combination of maturation and fertilization medium (18.45%). Therefore, it can be concluded that adding 1.0 mM glutathione in the maturation medium is more effective in increasing the competence of sheep oocytes to mature and results in improved fertilization rates. |
