| dc.description.abstract | Bleomisin digunakan secara luas sebagai antitumor, namun memiliki efek samping serius yaitu dapat menimbulkan stres oksidatif dan fibrosis yang dapat termanifestasi di berbagai organ tubuh termasuk limpa. Ciplukan (Physalis angulata) dapat menjadi alternatif pencegahan efek samping bleomisin karena mempunyai aktivitas terapeutik seperti antiinflamasi, antifibrosis, dan imunomodulator. Penelitian ini bertujuan untuk mengetahui dosis optimal dan efek herba ciplukan terhadap limpa tikus yang diinduksi bleomisin secara histopatologi. Sebanyak 21 ekor tikus dibagi menjadi 7 kelompok dengan masing-masing kelompok terdiri atas 3 ekor tikus kemudian dibedakan menjadi kelompok perlakuan tahap 14 dan 51 hari. Kelompok perlakuan terbagi menjadi kelompok kontrol negatif, bleomisin, serta bleomisin dan ciplukan (dosis 50 mg/kg BB dan 100 mg/kg BB). Evaluasi dilakukan dengan mengidentifikasi perubahan histopatologi limpa tikus dan skor kerusakan limpa dianalisis menggunakan uji Kruskal-Wallis dan post hoc dilakukan dengan uji Dunn. Kerusakan limpa oleh bleomisin ditunjukkan oleh temuan vasodilatasi, edema, deplesi pulpa putih, fibrosis, nekrosis, dan infiltrasi sel radang. Dosis optimal yang diperoleh adalah 50 mg/kg BB yang memiliki kemampuan untuk menekan kerusakan limpa, dibuktikan dengan perolehan skor yang paling mendekati kontrol negatif dan turunnya lesi fibrosis secara signifikan. | - |
| dc.description.abstract | Bleomycin is widely used for antitumor, however it has serious side effects by inducing oxidative stress and fibrosis that manifested in various organs including the spleen. Ciplukan (Physalis angulata) can be an alternative to prevent the side effects of bleomycin due to its therapeutic activities such as antiinflammatory, antifibrotic, and immunomodulatory. This research aims to determine the optimal dose and the effect of ciplukan on the spleen of rat induced by bleomycin through histopathology analysis. Total 21 rats were divided into 7 groups with each group consist of 3 rats then divided into 14 and 51-day-stage. The treatment groups were divided into negative control, bleomycin, and bleomycin combined with ciplukan (dose 50 mg/kg BW and 100 mg/kg BW). The evaluation performed by identifying the histopathological changes on rat’s spleen and scoring system were analyzed with Kruskal-Wallis test and post hoc Dunn's test. Spleen damages caused by bleomycin was indicated by findings of vasodilation, edema, white pulp depletion, fibrosis, necrosis, and inflammatory cell infiltration. The optimal dose obtained was 50 mg/kg BW, which had the ability to suppress spleen damage, as evidenced by the scores that were closest to the negative control and the fibrotic lesions decreased significantly. | - |
