| Abstract: | Kanker telah menjadi penyebab kematian terbesar di seluruh dunia, termasuk di Indonesia. Temu Ireng merupakan salah satu tanaman herbal yang berpotensi dikembangkan sebagai antikanker, karena kemampuannya dalam menginduksi apoptosis. Proses fermentasi menggunakan Aspergillus oryzae diharapkan dapat meningkatkan kandungan senyawa bioaktif ekstrak temu ireng. Penelitian ini bertujuan mengevaluasi kemampuan ekstrak temu ireng hasil fermentasi dan non fermentasi dalam menginduksi apoptosis. Penelitian ini menggunakan sel ragi sebagai model sel mamalia. Glukosa 4% digunakan sebagai kontrol positif, DMSO 1% sebagai kontrol negatif, dan metanol 2% sebagai kontrol pelarut. Uji induksi apoptosis dilakukan dengan parameter frekuensi sel petite dan aktivitas mitokondria pascainduksi. Hasil perhitungan frekuensi petite menunjukkan frekuensi sel petite tertinggi pada konsentrasi 12,5 ppm untuk ekstrak temu ireng hasil fermentasi dan non fermentasi yaitu sebesar 45% dan 67,07%. Pengujian aktivitas mitokondria menggunakan mikroskop konfokal dan rhodamin B sebagai pewarna menghasilkan penurunan intensitas pendaran warna merah seiring dengan penurunan konsentrasi ekstrak temu ireng. Cancer has become one of the leading causes of death worldwide, including in Indonesia. Curcuma aeruginosa (temu ireng) is a medicinal plant with potential to be developed as an anticancer agent due to its ability to induce apoptosis. Fermentation using Aspergillus oryzae is expected to enhance the bioactive compound content of C. aeruginosa extract. This study aimed to evaluate the apoptosis-inducing activity of fermented and non-fermented C. aeruginosa extracts. Yeast cells were used as a model for mammalian cells. A 4% glucose solution was used as a positive control, 1% DMSO as a negative control, and 2% methanol as a solvent control. Apoptosis induction was assessed based on petite cell frequency and mitochondrial activity post-induction. The highest petite cell frequencies were observed at a concentration of 12.5 ppm for both fermented and non-fermented extracts, at 45% and 67.07%, respectively. Mitochondrial activity tested using confocal microscopy and rhodamine B staining showed a decrease in red fluorescence intensity in line with increasing concentrations of C. aeruginosa extract. |
