| Abstract: | Perubahan iklim menyebabkan ancaman serius terhadap pertanian global, terutama karena meningkatnya suhu dan semakin sering terjadinya kekeringan. Hotong (Setaria italica L. Beauv.) memiliki metabolisme C4 yang adaptif terhadap perubahan iklim karena toleran terhadap kondisi lingkungan sub-optimum seperti kekeringan, tanah miskin hara, dan suhu tinggi. Karakterisasi genotipe hotong lokal Indonesia menjadi upaya awal dalam konservasi dan pengelolaan plasma nutfah Indonesia, dan juga merupakan tahapan penting dalam program pemuliaan untuk menghasilkan varietas unggul hotong. Penelitian ini bertujuan mengevaluasi keragaman genetik hotong lokal Indonesia menggunakan marka molekuler RAPD dan SCoT, dan mengidentifikasi amplikon spesifik genotipe menggunakan marka molekuler RAPD dan SCoT. Penelitian ini menggunakan 11 genotipe hotong lokal Indonesia dan menggunakan masing-masing 12 primer RAPD dan 12 primer SCoT sebagai marka dominan berbasis PCR. Sebanyak 104 total lokus terdeteksi menggunakan marka RAPD dengan rata-rata persentase polimorfisme 85,6% dan 121 total lokus dari marka SCoT dengan rata-rata persentase polimorfisme 94,2%. Lima primer RAPD dan tujuh primer SCoT menunjukkan persentase polimorfisme 100%. Rata-rata nilai polymorphism information content (PIC) untuk RAPD adalah 0,36 dan SCoT 0,37 menunjukkan marka RAPD dan SCoT yang digunakan cukup informatif. Analisis filogenik dalam bentuk konstruksi dendrogram membagi 11 genotipe menjadi dua kelompok besar yang memisahkan genotipe Botok-10 dengan genotipe lainnya. Sejumlah lokus spesifik genotipe berhasil diindentifikasi dari delapan genotipe hotong dan berpotensi dikembangkan menjadi marka sequence characterized amplified regions (SCAR). Climate change threatens global agriculture, mainly due to rising temperatures and increasing drought frequency. Foxtail millet (Setaria italica L. Beauv.) possesses a C4 metabolism that is adaptive to climate change, as it is tolerant to sub-optimal environmental conditions such as drought, poor soil fertility, and high temperatures. Characterizing Indonesian local foxtail millet genotypes is an initial effort to conserve and manage Indonesian germplasm. It is also a crucial step in breeding programs to produce superior foxtail millet varieties. This study aimed to evaluate the genetic diversity of local Indonesian foxtail millet using RAPD and SCoT molecular markers, and identify genotype-specific amplicons using RAPD and SCoT molecular markers. The study employed 11 local Indonesian foxtail millet genotypes and 12 RAPD and 12 SCoT primers as PCR-based dominant markers. A total of 104 loci were detected using RAPD markers with an average polymorphism percentage of 85.6%, and 121 total loci from SCoT markers with an average polymorphism percentage of 94.2%. Five RAPD primers and seven SCoT primers showed a 100% polymorphism percentage. The average polymorphism information content (PIC) values for RAPD and SCoT were 0.36 and 0.37, respectively, indicating that the RAPD and SCoT markers used were informative. Phylogenetic analysis in a dendrogram divided the 11 genotypes into two major groups, separating the Botok-10 genotype from the others. Several specific genotype loci were successfully identified from eight foxtail millet genotypes and could be developed into sequence-characterized amplified regions (SCAR) markers. |
