| Abstract: | Ketersediaan varietas tahan HLB menjadi kendala dalam pengembangan jeruk. Peningkatan produksi jeruk dapat dilakukan dengan pengembangan varietas baru tahan HLB yang telah dilakukan di BB BIOGEN sejak tahun 2018 melalui pemuliaan in vitro, penelitian tersebut telah menghasilkan beberapa klon jeruk yang terseleksi HLB melalui seleksi ketahanan secara in vitro. Penelitian ini bertujuan untuk mengkarakterisasi morfologi dan sitologi serta evaluasi ketahanan terhadap HLB dari keturunan klon-klon jeruk hasil pemuliaan in vitro yang terseleksi HLB. Pengamatan morfologi daun dilakukan terhadap ukuran dan bentuk daun, tinggi tanaman, diameter batang, dan kepadatan ranting. Pengamatan sitologi dilakukan terhadap panjang, lebar dan densitas stomata. Analisis klaster berdasarkan karakter morfologi dan sitologi menunjukkan pengelompokan yang mengacu pada asal-usul teknik pemuliaan yang digunakan, kecuali klon TR yang tidak mengelompok bersama klon FS. Hasil uji ketahanan HLB pada keturunan klon hasil pemulian menunjukkan terdapat 6 klon jeruk yang keturunannya tetap tahan yaitu, TR 79 450, TR 17 MV INV, FS 34 SOE, JCK 4C-2, JCK 3B-2, dan JC 500 11 C-2. The availability of HLB-resistant varieties is an obstacle in citrus development. Increasing citrus production can be done by developing new HLB-resistant varieties that have been carried out at BB BIOGEN since 2018 through in vitro breeding, the research has produced several HLB-selected citrus clones through in vitro resistance selection. This study aims to characterize the morphology and cytology as well as evaluate the resistance to HLB of the offspring of HLB-selected citrus clones from in vitro breeding. Morphological observations were made on leaf size and shape, plant height, stem diameter, and twig density. Cytological observations were made on the length, width and density of stomata. Cluster analysis based on morphological and cytological characters showed groupings that referred to the origin of the breeding techniques used, except for TR clones which did not cluster with FS clones. The results of the HLB resistance test on the offspring of the cultivated clones showed that there were 6 citrus clones whose offspring remained resistant, namely, TR 79 450, TR 17 MV INV, FS 34 SOE, JCK 4C-2, JCK 3B-2, and JC 500 11 C-2. |
