| Abstract: | Comminuted fracture merupakan jenis fraktur yang sulit ditangani karena tulang patah menjadi beberapa bagian kecil dan sering terdapat bagian tulang yang rusak atau hilang sehingga tulang tidak utuh. Scaffold demineralized freeze-dried bone xenograft (DFDBX) dan membran perikardium adalah bahan implan yang dapat digunakan sebagai penutup defect tulang. Secara klinis, penelitian kedua bahan tersebut belum banyak dilakukan pada hewan. Pengujian klinis dilakukan untuk memprediksi biokompatibilitas suatu bahan sehingga dapat digunakan pada hewan.
Penelitian ini bertujuan untuk mengevaluasi hemogram, radiodensitas dan ekhogenitas scaffold DFDBX dengan dan tanpa membran perikardium terhadap persembuhan defek tulang femur pada kucing. Sebanyak 15 ekor kucing jantan sehat berusia ±1 tahun, berat 2-3 kg dibagi menjadi tiga kelompok perlakuan. Tindakan bedah secara steril dan aseptis diawali dengan pembuatan defek tulang 3 mm pada os femur, kemudian diberi scaffold bone graft. Kelompok I adalah kontrol tanpa scaffold, kelompok II diberi scaffold DFDBX dan kelompok III diberi scaffold DFDBX+MP. Pemeriksaan darah, radiografi dan ultrasonografi dilakukan pada hari ke-0 sebelum perlakuan dan hari ke-1, 7, 14, dan 30 setelah perlakuan. Data diolah dengan menggunakan software excel, ImageJ® dan SPSS.
Hasil penelitian menunjukkan bahwa profil hematologi eritrosit, hemoglobin, hematokrit, MCV, MCHC kelompok DFDBX+MP mengalami penurunan secara signifikan (P<0.05) pada hari ke-14 dan 30 setelah perlakuan, platelet kelompok kontrol dan monosit kelompok DFDBX+MP mengalami peningkatan secara signifikan (P<0.05) pada hari ke-14 dan 30 setelah perlakuan. Analisis radiodensitas dan ekhogenitas setiap kelompok perlakuan pada tulang, peri-implan dan implan menunjukkan perbedaan yang tidak signifikan (P>0.05) selama 30 hari pengamatan dan kelompok DFDBX+MP memiliki tingkat persembuhan tulang yang lebih baik. Scaffold DFDBX dan DFDBX+MP dapat digunakan sebagai implan pada defek tulang. Comminuted fracture is a type of fracture that is complicated to handle due to the bone is broken into small fragments and is often found with defected bone so the bone is not intact. Scaffold demineralized freeze-dried bone xenograft (DFDBX) and pericardium membrane are implant materials that are able to use as lid for defect bone. Clinically, the study using those two materials were not much done before on animals. Clinical study that was done is to predict biocompatibility of a material so that is able to use on animal. This study aims to evaluate hemogram, radiodensity, and echogenicity of scaffold DFDBX with and without pericardium membrane to healing phase of defect of femoral bone of cat. As total of fifteen healthy male cats of aged 1-year-old and weighed 2-3 kg were divided into three treatment groups. Aseptic and sterile surgery was started by making of defect on 3 mm bone of femoral bone and was given by scaffold bone graft. Group I as a control group without scaffold, group II was given scaffold DFDBX, and group III was given scaffold DFDBX+MP. Blood examination, radiography, and ultrasonography were done on day-0 before treatment, and day-1, 7, 14, and 30 after treatment. Data was processed using software Microsoft Excel, ImageJ, and SPSS. The results showed that the hematological profile of erythrocytes, hemoglobin, hematocrit, MCV, MCHC of the DFDBX+MP group decrease significant change (P<0.05) on 14 and 30 days after treatment, platelets of the control group and monocytes of the DFDBX+MP group increase significant change (P<0.05) on 14 and 30 day after treatment. Analysis of radiodensity and echogenicity of every treatment group on bone, peri-implant, and implant showed not significant change (P>0.05) during 30 days of observation and group of DFDBX+MP had better bone healing rate. Scaffold DFDBX and DFDBX+MP are able to use as implants on defect bone. |
