Abstract: Kualitas sperma merupakan faktor utama yang menentukan keberhasilan inseminasi buatan dalam program pemuliaan ternak. Studi ini bertujuan untuk menganalisis hubungan antara kualitas sperma dan Parameter kinematika pada sperma kerbau murrah dan silangit dengan menggunakan computer-assisted sperm analysis (CASA). Sampel yang digunakan tiga pejantan dari masing-masing breed dengan tiga batch yang berbeda. Parameter kualitas sperma yang dievaluasi meliputi motilitas, viabilitas, abnormalitas, dan integritas membran plasma, sedangkan analisis kinematika mencakup kecepatan gerak (VAP, VSL, VCL) dan panjang lintasan (DAP, DSL, DCL) yang dianalisis dengan rancangan tersarang kemudian dikorelasikan dengan Peason Correlation. Hasil penelitian menunjukkan bahwa motilitas dan viabilitas sperma memiliki korelasi positif yang signifikan dengan parameter kinematika utama, yang mendukung peningkatan potensi fertilisasi. Sebaliknya, tingginya abnormalitas sperma berhubungan negatif dengan efisiensi pergerakan. Temuan ini dapat dijadikan dasar seleksi pejantan unggul guna meningkatkan tingkat keberhasilan inseminasi buatan. Kata kunci: inseminasi buatan, kerbau, kinematika, motilitas, sperm
Sperm quality is a critical factor determining the success of artificial insemination in livestock breeding programs. This study aimed to analyze the relationship between sperm quality and kinematic attributes in Murrah and Silangit buffalo sperm using Computer-Assisted Sperm Analysis (CASA). Samples were obtained from three bulls of each breed, collected over three different batches. The evaluated sperm quality parameters included motility, viability, abnormalities, and plasma membrane integrity, while the kinematic analysis focused on movement speeds (VAP, VSL, VCL) and path distances (DAP, DSL, DCL) using a nested design, which were subsequently correlated using Pearson Correlation. The results revealed that sperm motility and viability were significantly positively correlated with key kinematic parameters, supporting enhanced fertilization potential. Conversely, higher sperm abnormalities were negatively associated with movement efficiency. These findings can serve as a basis for selecting superior bulls to improve the success rate of artificial insemination.
