Abstract: Ekstrak bajakah (Uncaria sp.) telah terbukti mampu menginduksi apoptosis pada beberapa sel kanker, namun belum diketahui secara rinci mengenai proses induksi apoptosis tersebut. Penelitian ini bertujuan menentukan kemampuan ekstrak etanol kayu bajakah untuk menginduksi apoptosis pada sel ragi sebagai model sel mamalia. Ekstraksi dilakukan dengan metode maserasi, diikuti dengan uji fitokimia kualitatif, penentuan kadar fenolik dan flavonoid total. Selanjutnya, dilakukan uji induksi apoptosis dengan parameter frekuensi sel petite dan aktivitas mitokondria pascainduksi. Hasil positif diperoleh pada seluruh uji fitokimia kualitatif. Kadar fenolik total ekstrak yang diperoleh sebesar 27,665 mg GAE/g BK, sedangkan kadar flavonoid totalnya sebesar 8,711 mg QE/g BK. Hasil perhitungan frekuensi sel petite pascainduksi apoptosis menunjukkan frekuensi sel petite tertinggi pada konsentrasi 200 ppm, sebesar 91,35%, dan terendah pada konsentrasi 20 ppm, yaitu 5,32%. Penentuan aktivitas mitokondria sel yang diinduksi apoptosis menghasilkan penurunan intensitas pendaran warna merah seiring dengan peningkatan konsentrasi ekstrak bajakah
Bajakah (Uncaria sp.) extract can induce apoptosis in several cancer cells, yet its detailed mechanism remains unclear. This research aims to determine the ability of ethanol extract from bajakah wood to induce apoptosis in yeast cells, serving as a model for mammalian cells. Extraction was carried out with maceration method, followed by qualitative phytochemical screening, and determination of total phenolic and flavonoid content. Apoptosis induction was evaluated by petite cell frequency and mitochondrial activity post-induction. Positive results were seen in all phytochemical tests. The total phenolic content of the extract was 27.665 mg GAE/g DW, while the total flavonoid content was 8.711 mg QE/g DW. The results of the petite cell frequency count showed the highest petite cell frequency at 200 ppm, reaching 91.35%, and the lowest at 20 ppm, at 5.32%. The observation of mitochondrial activity in cells undergoing apoptosis showed a decrease in red fluorescence intensity as the concentration of the bajakah extract increased
