Abstract: Terapi berbasis sel mengalami perkembangan pesat dan banyak diteliti sebagai upaya pengobatan regenerasi jaringan atau organ yang rusak akibat kecelakaan fisik maupun bawaan, terapi penyakit degeneratif dan autoimun, serta terapi kecantikan. Mesenchymal stem cells (MSC) termasuk sel punca yang memiliki sifat immunomodulasi yang berperan terhadap proses penyembuhan regenerasi. Umbilical cord Wharton jelly mengekspresikan sifat dan karakteristik dari MSC. Penelitian ini bertujuan mengisolasi Wharton jelly pada umbilical cord sebagai eksplan untuk memperoleh MSC dan menganalisis karakter imunofenotipenya. Metode yang digunakan meliputi isolasi secara eksplan umbilical cord dari persalinan sectio caesarian dengan mengkultur jaringan Wharton jelly untuk menumbuhkan dan memperbanyak MSC, serta menganalisis proses tumbuhnya MSC dan total sel selama pasase. Analisis imunofenotipe menggunakan flow cytometry dalam identifikasi Cluster of Differentiation (CD) berupa CD73, CD90, dan CD 105 sebagai ciri dari MSC. Spourting atau tumbuhnya MSC terjadi pada hari ke-3 setelah isolasi. Pada hari ke-7, morfologi sel dapat terlihat sedikit lebih jelas berbentuk fibroblast like cells. Rerata total sel pada pasase 1, 2, 3, dan 4 adalah 141.250, 445.000, 907.500, 1.608.750 sel. Imunofenotipe sel kultur MSC dari Wharton jelly pada pasase 4 memiliki nilai persentase biomarker protein membran MSC dengan CD73 (99.3%), CD90 (99.8%), dan CD105 (94.7%) yang mengindikasikan sel hasil kultur berupa MSC.
Cells based therapies have developed rapidly and are widely researched as a solution to the treatment of tissue or organ regeneration damaged by physical or congenital accidents, therapy for degenerative and autoimmune diseases, and beauty therapy. Mesenchymal stem cells (MSC) include stem cells that have immunomodulating properties that play a role in the regeneration healing process. Umbilical cord Wharton jelly expresses the properties and characteristics of MSC. This study aims to isolate Wharton jelly in umbilical cord as an explant to obtain MSCs and analyze its immonufenotype characters. The method used includes explant isolation of umbilical cord from sectio caesarian delivery by culturing Wharton jelly tissue to grow and multiply MSC and analyzing the process of MSC growth and total cells during passage. Immunophenotype analysis using flow cytometry in identifying Cluster of Differentiation (CD) in CD73, CD90, and CD 105 as a characteristic of MSC. Spourting or growth of MSCs occurred on day 3 after isolation. On day 7, cells morphology can be seen more clearly in the form of fibroblast like cells. The average total cells in passage 1, 2, 3, and 4 were 141,250, 445,000, 907,500, 1,608,750 cells. Immunophenotype of MSC culture cells from Wharton jelly at passage 4 has a percentage value of MSC membrane protein biomarkers with CD73 (99.3%), CD90 (99.8%), and CD105 (94.7%) which indicates the cultured cells are MSC.
