dc.description.abstractArcangelisia flava, atau "kayu kuning", adalah tumbuhan obat yang digunakan untuk mengobati berbagai penyakit termasuk diabetes melitus tipe 2 (T2DM). Meskipun banyak digunakan, mekanisme farmakologis A. flava dalam pengobatan T2DM belum diketahui secara pasti. Penelitian ini bertujuan untuk menganalisis potensi A. flava sebagai obat oral T2DM dengan menggunakan jejaring farmakologi dan penambatan molekuler. Dari 47 metabolit sekunder yang diidentifikasi, 23 diantaranya memenuhi parameter absorpsi obat. Melalui jejaring farmakologi dan interaksi protein-protein, diidentifikasi kandidat potensial untuk pengobatan T2DM, yaitu 3-hydroxy-3',4',5'-trimethoxyflavone, 8-hydroxy-berberine, berberrubine, columbamine, dehydrocorydalmine, demethyleneberberine, dihydroberberine, jatrorrhizine, pallidinine, dan palmatrubine. Target protein potensial yang diidentifikasi antara lain AKT1, TNF, BCL2, CASP3, EGFR, PPARG, dan SIRT1. Mekanisme A. flava dalam pengobatan T2DM diprediksi berkaitan dengan jalur lintasan toksoplasmosis, penyakit kanker, resistensi insulin, apoptosis, serta jalur lintasan pensinyalan yang berhubungan dengan diabetes (AGE-RAGE, MAPK, PI3K-Akt, adipositokin, dan insulin). Nilai afinitas pengikatan antara ligan-reseptor kandidat menunjukkan interaksi yang kuat, dengan nilai berkisar antara -5,5 hingga -9 kkal/mol. Hasil dari penelitian ini menunjukkan bahwa A. flava berpotensi sebagai obat alternatif untuk T2DM dengan mekanisme yang melibatkan berbagai jalur sinyal penting terkait diabetes, kanker, dan toksoplasmosis. Penelitian memberikan informasi mengenai A. flava yang dapat diteliti dan dikembangkan lebih lanjut sebagai obat oral T2DM.
dc.description.abstractArcangelisia flava, or "yellow fruit moonseed", is a medicinal plant used to treat various diseases, including type 2 diabetes mellitus (T2DM). Despite its widespread use, the pharmacological mechanisms of A. flava in treating T2DM remain unclear. This study aimed to analyze the potential of A. flava as an oral T2DM drug using pharmacological network and molecular docking analysis. Among the 47 identified secondary metabolites, 23 metabolites met drug absorption parameters. Through pharmacological network and protein-protein interaction analysis, potential candidates for T2DM treatment were identified, such as 3-hydroxy-3',4',5'-trimethoxyflavone, 8-hydroxy-berberine, berberrubine, columbamine, dehydrocorydalmine, demethyleneberberine, dihydroberberine, jatrorrhizine, pallidinine, and palmatrubine. The potential target proteins identified are AKT1, TNF, BCL2, CASP3, EGFR, PPARG, and SIRT1. The mechanisms of A. flava in T2DM treatment likely involve pathways related to toxoplasmosis, cancer, insulin resistance, apoptosis, and diabetes-related signaling pathways (AGE-RAGE, MAPK, PI3K-Akt, adipocytokine, and insulin). The binding affinity values between candidate ligands and receptors indicate strong interactions, with values ranging from -5.5 to -9 kcal/mol. These results suggest that A. flava has potential as an alternative treatment for T2DM, with mechanisms involving various key signaling pathways related to diabetes, cancer, and toxoplasmosis. This research supports further exploration of A. flava as an oral T2DM drug.
