| Abstract: | Swab permukaan merupakan metode paling umum yang digunakan untuk memantau kontaminasi patogen pada permukaan dan menentukan cukupnya kinerja pembersihan. Menurut ISO 18593:2018, penundaan waktu antara pengambilan sampel swab permukaan dan analisis harus dilakukan sesingkat mungkin. Namun, pada kenyataannya, dalam industri terkadang terjadi penundaan atau keterlambatan analisis. Penelitian ini bertujuan untuk mengetahui pengaruh waktu penyimpanan terhadap pertumbuhan bakteri dan jamur. Penelitian dilakukan dengan cara menambahkan spike bakteri/jamur kedalam media enumerasi yang disimpan pada suhu 1–8 °C selama tujuh hari, kemudian dilakukan perhitungan jumlah bakteri/jamur yang tumbuh setiap interval 24 jam. Berdasarkan hasil analisis diperoleh bahwa pertumbuhan bakteri dan jamur mengalami fase adaptasi, pertumbuhan sampai titik optimum, dan penurunan seiring lamanya penyimpanan sampel. Fase adaptasi bakteri dan jamur ini akan menjadi batas optimum penyimpanan sampel swab oles permukaan yaitu dengan batas penyimpanan selama empat hari untuk memastikan hasilnya tetap mewakili kondisi aktual. Surface swabbing is the most common method used to monitor pathogen contamination on surfaces and determine cleaning performance adequacy. According to ISO 18593:2018, the delay between surface swab sampling and analysis should be minimized. However, delays or postponements in analysis can occur in the industry. This study aims to determine the effect of storage time on bacterial and fungal growth and establish the duration for which swab samples can be stored while still providing representative results. The research involved adding bacterial/fungal spikes into the enumeration medium (sterile water), storing them at 1–8 °C for 7 days, and counting the bacteria/fungi every 24 hours. Initially, bacterial and fungal growth did not occur due to the adaptation phase. After adaptation, the number of bacteria and fungi increased with longer storage time until reaching an optimum point, after which growth declined. The study concluded that surface swab samples should only be stored for 4 days for both fungal and bacterial parameters to ensure results remain representative of actual conditions. |
