Abstract: Kumis kucing (Orthosiphon aristatus) merupakan tanaman herbal yang memiliki senyawa polifenol sehingga berpotensi sebagai antioksidan. Penelitian ini bertujuan menentukan kombinasi jenis dan rasio pelarut terbaik untuk mengekstrak daun kumis kucing secara Microwave Assisted Extraction (MAE) berdasarkan kadar total polifenol dan kapasitas antioksidan menggunakan Response Surface Methodology (RSM). Ekstraksi dilakukan menggunakan one factorial dengan diawali oleh pelarut (aseton, etanol, metanol, akuades) dan rasio (3:1, 5:1, 10:1, 15:1). Data hasil optimasi menujukkan total fenolik tertinggi pada konsentrasi aseton 30% dan rasio 3:1 sebesar 58,21 mg GAE/g DW. Total flavonoid tertinggi pada konsentrasi aseton 80% dan rasio 4,96:1 sebesar 9,71 mg QE/g DW. Kapasitas FRAP tertinggi pada konsentrasi aseton 46,5% dan rasio 3:1 sebesar 600,67 µmol TE/g DW, sedangkan DPPH tertinggi pada konsentrasi aseton 59% dan rasio 4,1:1 sebesar 1,03 µmol TE/g DW. Solusi optimasi pelarut yang didapat yaitu dengan penggunaan konsentrasi aseton 96% dan rasio 2:1, dengan desirability sebesar 0,491.
Cat's whiskers (Orthosiphon aristatus) were a herbal plant that contained polyphenolic compounds so it had the potential to act as an antioxidant. This research aimed to determine the best solvent combination and solvent to solid ratio to extract cat's whisker leaves using Microwave Assisted Extraction (MAE) based on total polyphenol content and antioxidant capacity using Response Surface Methodology (RSM). Extraction was carried out using one factorial starting with solvent (acetone, ethanol, methanol, aquades) and ratio (3:1, 5:1, 10:1, 15:1). Optimization data showed the highest total phenolics at an acetone concentration of 30% and a 3:1 ratio of 58,21 mg GAE/g DW. The highest total flavonoids were at an acetone concentration of 80% and a ratio of 4.96:1 of 9,71 mg QE/g DW. The highest FRAP capacity was at an acetone concentration of 46.5% and a 3:1 ratio of 600,67 µmol TE/g DW, while the highest DPPH was at an acetone concentration of 59% and a 4,1:1 ratio of 1,03 µmol TE/g DW. The solvent optimization solution obtained was by using an acetone concentration of 96% and a ratio of 2:1, with a desirability of 0.491.
