Abstract: PT X merupakan perusahaan swasta nasional yang memproduksi susu berprotein tinggi. PT X menggunakan isolat protein kedelai (IPK) sebagai bahan baku yang akan meningkatkan kandungan protein dalam produknya. Oleh karena itu, kadar protein dalam IPK harus diperhatikan agar memberikan hasil yang baik. PT X memiliki laboratorium internal yang menguji kadar protein dengan metode modifikasi SNI 01-2891-1992. Namun hasil uji kadar protein dalam IPK dengan metode tersebut belum mencapai kadar yang sesuai dengan CoA. Hasil penelitian didapatkan metode dengan kondisi paling optimal diperoleh mengacu pada metode “supplier” dengan optimasi penggunaan katalis selenium, volume H2SO4 12 mL, dan volume NaOH 60 mL. Metode ini divalidasi dengan parameter presisi dan robustness. Uji presisi menghasilkan %RSD 0,81 < 2/3 CV Horwitz-nya 5,41%, nilai ini menunjukan metode dapat dikatakan presisi. Uji robustness dilakukan dengan alternatif perlakuan penggunaan volume H2SO4 11 dan 13 mL, didapat Fhitung <Ftabel dan thitung <ttabel, hasil ini menunjukkan metode robust terhadap alternatif perlakuan penggunaan volume H2SO4 11 dan 13 mL.
PT X is a national private company that produces high-protein milk. PT X uses isolate soy protein (ISP) as a raw material that will increase the protein content in its products. Therefore, the protein content in ISP must be considered in order to provide good results. PT X has an internal laboratory that tests protein content using a modified method of determining crude protein content according to SNI 01-2891-1992. However, the results of the protein content test in IPK with this method have not reached the level that is in accordance with CoA. The results showed that the method with the most optimal conditions obtained refers to the “supplier” method with optimization of the use of selenium catalyst, H2SO4 volume of 12 mL, and NaOH volume of 60 mL. The method was validated with precision and robustness parameters. The precision test resulted in a %RSD of 0.81 < 2/3 of CV Horwitz of 5.41%, this value indicates the method can be said to be precise. Robustness test was conducted with alternative treatment of the use of 11 and 13 mL H2SO4 volume, obtained Fstat< Ftabel and tstat<ttabel, these results indicate the method is robust to alternative treatment.
