| dc.description.abstract | Lepidoptera predator yang diduga berpotensi untuk dikembangkan sebagai agens pengendali hayati, ditemukan memangsa kutu tempurung (Coccidae) pada tanaman kopi (Coffea spp.) di Sukabumi, Jawa Barat. Namun demikian, belum banyak informasi yang diketahui mengenai predator tersebut di Indonesia. Oleh karena itu, penelitian dilakukan untuk mengetahui spesies predator, mendapat informasi biologi, serta untuk menganalisis potensi pemangsaan larva predator terhadap hama kutu tempurung yang menyerang tanaman kopi. Penelitian dilaksanakan di Sukabumi dan Bogor, mulai bulan Juni 2022 sampai November 2023. Predator diidentifikasi secara molekuler di laboratorium untuk mengetahui spesiesnya menggunakan prosedur PCR terhadap gen COI mitokondria. Identifikasi molekuler terdiri atas ekstraksi DNA, amplifikasi dengan primer universal LCO-HCO, elektroforesis, sequencing, dan analisis data. Analisis data dilakukan berdasarkan penjajaran sekuen DNA dan homologi. Larva predator dikumpulkan dari kebun kopi di Sukabumi, kemudian dipelihara di rumah kaca dengan Coccus sp. sebagai pakan. Perbanyakan Coccus sp. dilakukan pada bibit kopi. Selanjutnya, pengamatan dilakukan terhadap perkembangan dan kemampuan pemangsaan predator. Hasil identifikasi molekuler menunjukkan bahwa sekuen DNA sampel (658 bp) mempunyai tingkat kemiripan tertinggi sebesar 95,44% (GenBank) terhadap spesies Autoba rubra. Dengan demikian dapat disimpulkan bahwa Lepidoptera predator yang memangsa kutu tempurung pada tanaman kopi di Sukabumi adalah A. rubra. Telur predator menetas dalam waktu 6,29 ± 0,59 hari di laboratorium. Lepidoptera predator diduga mengalami perkembangan empat instar larva dengan durasi keseluruhan 28,76 ± 4,89 hari di laboratorium. Tahap pupa berlangsung selama 21,00 ± 5,12 hari di laboratorium. Nisbah kelamin jantan:betina adalah 1:2,4 di laboratorium dan 1:1,02 di lapangan. Betina dan jantan masing-masing hidup selama 4,75 ± 1,29 dan 5,20 ± 0,84 hari di laboratorium dan 5,07 ± 1,13 dan 5,72 ± 1,36 hari di lapangan. Baik betina yang kawin maupun yang tidak kawin dapat bertelur. Betina yang kawin yang dikumpulkan dari lapang menghasilkan 0–183 butir telur, sedangkan yang dipelihara di laboratorium menghasilkan 5–9 butir telur. Betina yang tidak kawin, baik yang dikumpulkan dari lapang maupun yang dipelihara di laboratorium menghasilkan jumlah telur yang hampir sama yaitu 15,9 butir dan 14,87 butir masing-masing. Telur diletakkan secara sendiri-sendiri. Kemampuan pemangsaan predator terhadap Coccus sp. di laboratorium sebesar 138,17 ± 37,64 kutu/larva. Larva predator memangsa imago dan nimfa kutu. | |
| dc.description.abstract | The predatory lepidoptera, identified as a potential biological control agent for the soft scale insect (Coccidae) on coffee plants (Coffea spp.), was discovered in Sukabumi, West Java. However, information on this predatory lepidoptera, particularly in Indonesia, is limited. Therefore, this study aimed to identify the predator, gather biological information, and assess its capacity to control soft scale insects. The research was conducted in Sukabumi and Bogor from June 2022 to November 2023. The predator was identified molecularly in the laboratory using PCR procedures targeting the mitochondrial COI gene. The molecular identification process included DNA extraction, DNA amplification using LCO-HCO universal primers, electrophoresis, DNA sequencing, and data analysis. Data analysis involved DNA alignment and homology comparison. Predatory larvae were collected from coffee plantations in Sukabumi and then reared in a greenhouse, with Coccus sp. as their feed. The rearing of Coccus sp. was carried out on coffee seedlings. Observations focused on the predator's development and predation capacity. Molecular identification results showed that the sample DNA sequences (658 bp) had a 95.44% similarity to the species Autoba rubra according to GenBank. Thus, it was concluded that the predatory lepidoptera preying on soft scale insects on coffee plants in Sukabumi is A. rubra. The eggs of the predator hatched in 6.29 ± 0.59 days in the laboratory. The predator undergoes four larval instars with a total duration of 28.76 ± 4.89 days. The pupal stage lasts on average 21.00 ± 5.12 days. The sex ratio of males to females is 1:2.4 in the laboratory and 1:1.02 in the field. Female and male predators live for 4.75 ± 1.29 and 5.20 ± 0.84 days, respectively, in the laboratory, and 5.07 ± 1.13 and 5.72 ± 1.36 days, respectively, in the field. Both mated and unmated female predators lay eggs. Mated females collected from the field laid 0–183 eggs, while those reared in the laboratory laid 5–9 eggs. Unmated females laid an average of 15.9 eggs in the field and 14.87 eggs in the laboratory. The eggs were laid singly. The predator's capacity to prey on Coccus sp. in the laboratory is 138.17 ± 37.64 scales per larva. Predatory larvae feed on both adult and nymph stages of the scales. | |
