Abstract: Jenis tanaman yang umum digunakan di Hutan Tanaman Industri (HTI) sebagai bahan baku pulp dan kertas adalah Eucalyptus pellita. Bibit berkualitas E. pellita dapat diproduksi dari klon unggul dengan perbanyakan secara vegetatif, yakni stek pucuk (tunas). Perbanyakan tanaman melalui stek pada umumnya menggunakan hormon untuk membantu proses terbentuknya akar, namun beberapa klon E. pellita ada yang mudah berakar tanpa bantuan hormon. Oleh karena itu perlu dilakukan penelitian analisis fisiologi stek pucuk E. pellita (klon 148) tanpa menggunakan hormon. Desain penelitian yang digunakan adalah Rancangan Acak Lengkap dengan lima perlakuan yakni umur tunas 30, 27, 24, 21, dan 18 hari dengan 5 ulangan. Hasil analisis fisiologi stek pucuk E. pellita yakni kandungan hormon IAA, C-organik, dan N menunjukkan hasil yang tidak linear dengan perlakuan umur tunas. Hasil pengukuran panjang tunas, jumlah node, dan jarak antar node menunjukkan nilai yang linear dengan perlakuan umur tunas. Hasil pengamatan total hidup (survival rate), total berakar, dan bertunas menunjukkan perlakuan umur tunas 18 hari memiliki nilai terendah dibandingkan perlakuan lain. Berdasarkan hasil pengukuran tinggi dan diameter umur 12 MST menunjukkan perlakuan tunas umur 21 hari memiliki pertumbuhan dan survival rate terbaik sehingga dapat menjadi dasar penentuan umur tunas dalam produksi bibit klon 148 skala operasional. Eucalyptus pellita is extensively utilized in Industrial Plantation Forests (HTI) as a primary source for pulp and paper production. High-quality seedlings of E. pellita can be produced from superior clones through vegetative propagation, particularly using shoot cuttings. Although hormonal treatments are commonly applied to facilitate rooting, some E. pellita clones can root effectively without them. This study examines the physiological characteristics of E. pellita shoot cuttings (clone 148) without hormone application. The research employed a Completely Randomized Design with treatments corresponding to shoot ages of 30, 27, 24, 21, and 18 days, each replicated five times. Results showed non-linear relationships between shoot age and the levels of IAA hormone, organic carbon, and nitrogen. However, shoot length, number of nodes, and internode distance exhibited linear relationships with shoot age. The 18-day shoot treatment had the lowest survival rate, rooting, and sprouting. Measurements at 12 weeks post-planting indicated that 21-day-old shoots demonstrated superior growth and survival rates. These findings suggest that 21-day-old shoots are optimal for the operational-scale production of clone 148 seedlings. Key words: Eucalyptus pellita, hormones, physiology, propagation
