Abstract: Jenis kelamin merupakan informasi dasar dari makhluk hidup yang penting untuk diketahui, namun tidak semua makhluk hidup mudah untuk dibedakan antara jantan dan betinanya. Beberapa spesies memiliki penampakan morfologis yang sama antara jantan dan betinanya, sehingga sulit dibedakan contohnya pada burung. Pada dasarnya, telah tersedia berbagai metode macam dalam menentukan jenis kelamin burung. Rata-rata kemurnian DNA darah sebesar 1,425 yang berarti DNA yang dihasilkan tidak terlalu murni. Rata-rata kemurnian DNA bulu yaitu 1,526 yang berarti DNA yang dihasilkan berada dibawah kisaran DNA murni yaitu 1,8 – 2,0. Berdasarkan hasil PCR, sampel bulu dari burung dapat digunakan sebagai sumber DNA, karena memberikan hasil yang sama dengan DNA yang berasal dari darah. Hasil pengamatan gambaran struktur makroskopis ovarium pada burung merpati betina terlihat berbentuk bulat kecil, tidak terlihat pembuluh darah, berukuran kurang lebih 4-6 mm, serta berwarna putih susu. Hasil pengamatan gambaran struktur makroskopis testikel akan terlihat berbentuk bulat hingga oval berjumlah satu pasang dan ovarium akan ditemukan dengan bentuk bulat atau oval dan terdapat folikel-folikel.
Gender is basic information about living creatures that is important to know, but not all living creatures are easy to distinguish between males and females. Some species have the same morphological appearance between males and females, making it difficult to differentiate them, for example in birds. Basically, there are various methods available to determine the sex of a bird. The average purity of blood DNA is 1.425, which means the DNA produced is not very pure. The average purity of feather DNA is 1.526, which means the DNA produced is below the pure DNA range, namely 1.8 – 2.0. Based on PCR results, feather samples from birds can be used as a DNA source, because they give the same results as DNA from blood. The results of observations of the macroscopic structure of the ovaries in female pigeons show that they are small round in shape, with no blood vessels visible, measuring approximately 4-6 mm, and milky white in color. As a result of observing the macroscopic structure of the testicles, one pair will appear to be round to oval in shape and the ovaries will be found to be round or oval in shape and contain follicle
