Abstract: Masyarakat Melayu di Kabupaten Belitung dominan memanfaatkan tumbuhan obat dari famili Myrtaceae, seperti Syzygium bankense, Syzygium euneuron, dan Rhodomyrtus tomentosa. Namun, informasi metabolitdari ketiga spesies tersebut masih terbatas. Profil metabolit suatu tumbuhan dapat ditentukan dengan cepat melalui studi metabolomik. Penelitian ini bertujuan menganalisis metabolit tak tertarget daun S. bankense, S. euneuron, dan R. tomentosa dari Kabupaten Belitung dan menentukan penanda kimia ketiga spesies untuk konstruksi kemosistematika menggunakan pendekatan metabolomik berbasis liquid chromatography mass spectrometry (LC-MS/MS). Daun ketiga spesies dimaserasi menggunakan etanol, lalu ekstrak dianalisis dengan LCMS/MS. Identifikasi metabolit menggunakan MZmine dan analisis multivariat menggunakan MetaboAnalyst. Sebanyak 639 metabolit teridentifikasi dari spesies S. bankense (450), S. euneuron (452), dan R. tomentosa (451). Kelas metabolit yang teridentifikasi di ketiga spesies yaitu asam lemak (29), alkaloid (36), fenolik (21), flavonoid (52), kumarin (19), terpenoid (43), karbohidrat (33), dan asam amino (46). Metabolit kandidat penanda kimia untuk S. bankense berjumlah 11, S. euneuron berjumlah 22, dan R. tomentosa berjumlah 47. Penanda kimia dengan skor variable importance on projection (VIP) tertinggi yaitu asam vanililmandelat pada S. bankense, asam madekasik pada S. euneuron, dan skropoliosida D pada R. tomentosa. Berdasarkan profil metabolit, S. bankense lebih dekat dengan S. euneuron dibandingkan R. tomentosa.
The Malay Belitung community in Belitung Regency dominates the use of medicinal plants from the Mytaceae family, like Syzygium bankense, Syzygium euneuron, and Rhodomyrtus tomentosa. However, the metabolite information of leaves of the three species was unexplored. The metabolite profile of a plant can be determined rapidly through metabolomics studies.The study aims to analyze the metabolites of S. bankense, S. euneuron, and R. tomentosa leaves from the Belitung Regency and determine the markers of the species for the chemosystematic construction using a metabolomic approach based on liquid chromatography-mass spectrometry (LC-MS/MS). The leaves are macerated with ethanol, and extracts are analyzed with LC-MS/MS. Metabolite identification used MZmine and multivariate analysis was performed using MetaboAnalyst. Identified 639 metabolites of the species S. bankense (450), S. euneuron (452), and R. tomentosa (451). The metabolite classes identified in the three species are fatty acids (29), alkaloids (36), phenolic (21), flavonoids (52), coumarin (19), terpenoids (43), carbohydrates (33), and amino acids (46). The number of candidate metabolites markers are S. bankense (11), S. euneuron (22) and R. tomentosa (47), as well as the marker with the highest variable importance on projection (VIP) is vanillylmandelic acid in S. bankense, madeccasic acid in S. euneuron, and scropolioside D in R. tomentosa. Based on the metabolite profile, S. bankense is closer to S. euneuron than R. tomentosa.
