| dc.description.abstract | Haemonchosis merupakan penyakit gastrointestinal yang disebabkan infeksi cacing Haemonchus contortus pada domba. Pemeriksaan molekuler menggunakan metode PCR penting dilakukan untuk mengetahui tingkat patogenitas Haemonchus contortus. Namun, ukuran larva Haemonchus contortus yang kecil meningkatkan resiko tidak terambilnya larva saat proses preparasi. Diperlukan optimasi berbagai metode preparasi larva Haemonchus contortus untuk mendapatkan cara preparasi pemeriksaan molekuler DNA Haemonchus contortus dengan hasil terbaik. Tujuan penelitian ini adalah menguji metode preparasi larva Haemonchus contortus paling optimal untuk identifikasi molekuler. Metode preparasi sampel larva terdiri atas 5 kelompok berdasarkan perbedaan volume dan media larva menggunakan akuades dan PBS. Proses PCR terdiri atas tahap ekstraksi DNA dan pelaksanaan PCR. Produk PCR yang dibaca dengan elektroforesis menggunakan agarose 1% menunjukkan preparasi media akuades menghasilkan pita yang lebih terang dibandingkan preparasi dengan media PBS dengan panjang pita DNA sebesar 260 bp. Penggunaan akuades sebagai media pembawa larva pada kedua volume preparasi tidak memengaruhi hasil PCR secara signifikan sehingga akuades dapat digunakan sebagai media pembawa larva Haemonchus contortus yang berukuran kecil. | id |
| dc.description.abstract | Haemonchosis is a gastrointestinal disease caused by Haemonchus contortus worm infection in sheep. Molecular examination using the PCR method is important to determine the level of pathogenicity of Haemonchus contortus. However, the small size of Haemonchus contortus larvae increases the risk of not capturing the larvae during the preparation. Methods Optimization of preparation of Haemonchus contortus larvae is needed to obtain a preparation method for molecular examination of Haemonchus contortus DNA with the best results. The aim of this study was to obtain the most optimal Haemonchus contortus larval preparation method for molecular identification. The larval sample preparation method consisted of 5 groups based on differences in volume and larval media using aquades and PBS. The PCR process consists of DNA extraction and PCR implementation. PCR product were read by electrophoresis using 1% agarose, showing the preparation of distilled water media produced a brighter band than preparation with PBS media with a DNA band length of 260 bp. The use of aquades as a medium for larvae in both preparation volumes did not greatly affect the PCR results so aquades can be used as a medium for small Haemonchus contortus larvae. | id |
