Abstract: | Sorgum (Sorghum bicolor) adalah tanaman serealia terpenting kelima di
dunia setelah beras, gandum, jagung dan barli yang memiliki banyak manfaat antara
lain sebagai bahan pangan, pakan, dan industri. Sorgum toleran terhadap
kekeringan dan lingkungan marjinal sehingga baik untuk dikembangkan di
beberapa wilayah di Indonesia. Pengembangan tanaman sorgum menghadapi
beberapa kendala dan salah satunya adalah gangguan penyakit. Penyakit bercak ter
adalah salah satu penyakit pada tanaman sorgum yang disebabkan oleh infeksi
cendawan Phyllachora sp. Pengetahuan yang komprehensif tentang ekologi
patogen dan respons ketahanan tanaman sorgum terhadap infeksi Phyllachora
sangat diperlukan untuk pengembangan sorgum tahan penyakit bercak ter.
Meskipun penyakit ini berdampak kecil pada kehilangan hasil tapi pada tingkat
infeksi yang lanjut akan berdampak pada kehilangan hasil yang cukup besar.
Lignin sangat berperan dalam ketahanan tanaman terhadap infeksi patogen
sehingga kajian keterkaitan yang erat antara lignin dengan ketahanan tanaman
terhadap penyakit sangat diperlukan.
Penelitian dilakukan dengan tujuan (1) mendapatkan pengetahuan tentang
karakter morfologi dan molekuler cendawan Phyllachora sp. pada tanaman sorgum
yang ada di Indonesia; (2) mengetahui insidensi dan keparahan penyakit bercak ter
dari Bogor, Gunung Kidul dan Lombok Barat; (3) mendapatkan pengetahuan
tentang hubungan kandungan lignin dengan ketahanan tanaman sorgum terhadap
infeksi Phyllachora sp melalui uji evaluasi lapangan; (4) mengetahui komposisi
monolignol dan ekspresi gen biosintesis lignin pada daun tanaman sorgum sebagai
faktor yang mempengaruhi ketahanan tanaman sorgum terhadap penyakit bercak
ter. Identifikasi cendawan umumnya dilakukan melalui pendekatan morfologi dan
molekuler. Beberapa karakter morfologi cendawan Phyllachora sp. yang dapat
digunakan untuk identifikasi adalah karakter dan panjang parafisis, bentuk dan
ukuran askus serta askospora. Pendekatan molekuler sangat penting dalam
identifikasi Phyllachora yang merupakan cendawan obligat, karena sulit untuk
ditumbuhkan pada media kultur cendawan. Identifikasi molekuler cendawan
menggunakan metode polymerase chain reaction (PCR) dilakukan menggunakan
marka universal cendawan, yaitu marka yang mengamplifikasi daerah internal
transcribed spacer (ITS) dan large sub unit (LSU) kemudian dilanjutkan dengan
analisis sekuensing.
Respons ketahanan tanaman sorgum terhadap infeksi Phyllachora terkait
dengan kandungan lignin total pada tanaman sorgum dilakukan melalui uji evaluasi
lapangan. Mekanisme ketahanan tanaman yang didasarkan pada perubahan
komposisi dan biosintesis lignin dilakukan melalui analisis komposisi monolignol
pada daun sorgum dengan pendekatan metode Pyrolysis Gas Chromatography
Mass Spectrometry (Py-GCMS). Sementara analisis ekspresi gen penyandi
bosintesis lignin dilakukan dengan menggunakan marka gen SbCOMT dengan
pendekatan metode Real Time quantitative PCR (RT-qPCR). Selain itu,
pengamatan beberapa sifat agronomi yang berkaitan dengan biomasa (diantaranya
tinggi tanaman, diameter batang, panjang malai, bobot daun, bobot batang, bobot
malai dan bobot seratus biji) dilakukan untuk mempelajari hubungan kandungan
lignin dengan sifat agronomi masing-masing genotipe. Hal ini penting sebagai
dasar menentukan jenis genotipe sorgum yang akan ditanam sesuai fungsinya,
misalnya sebagai sumber bahan pangan, pakan atau biomaterial.
Penelitian ini menunjukkan nilai keparahan penyakit bercak ter yang teramati
bervariasi dari 32,4% pada genotipe Samurai yang rentan dan 87,9% pada genotipe
Latu Keta yang tahan terhadap penyakit bercak ter. Hasil identifikasi morfologi dari
17 genotipe menunjukkan karakteristik yang berbeda-beda sehingga dapat
dikelompokkan menjadi beberapa kelompok spesies dan diduga kuat adalah spesies
yang baru. Identifikasi molekuler dengan marka yang mengamplifikasi daerah ITS
dan LSU masing-masing menunjukkan dua isolat dan delapan isolate merupakan
cendawan Phyllachora. Hasil karakterisasi dengan morfologi dan molekuler
menunjukkan konsistensi yang melandasi pernyataan bahwa identifikasi dan
karakterisasi cendawan dengan pendekatan morfologi masih menjadi kunci dasar
indentifikasi dan karakterisasi cendawan yang diverifikasi dan dilengkapi dengan
pendekatan molekuler. Hasil penelitian ini juga menunjukkan keterkaitan
kandungan lignin total dengan ketahanan tananam terhadap penyakit bercak ter.
Hasil analisis komposisi monolignol memberikan hasil yang serupa dan konsisten
dengan hasil analisis ekspresi gen SbCOMT. Genotipe 4183A menunjukkan nilai
ketahanan yang tinggi terhadap penyakit bercak ter.
Hasil penelitian ini diharapkan dapat memberikan manfaat antara lain
(1) menjadi pengetahuan dasar bagi fitopatologis dalam mengkaji lebih jauh tentang
cendawan Phyllachora sp. penyebab penyakit bercak ter pada tanaman sorgum.
Manfaat lain adalah (2) menjadi pengetahuan yang berguna bagi pemuliaan
tanaman sorgum dalam pengembangan tanaman sorgum tahan terhadap penyakit
bercak ter. Manfaat terakir dari penelitian ini adalah (3) rekomendasi penggunaan
genotipe sorgum untuk berbagai kepentingan baik pangan, pakan dan industri
disertai saran teknik budi daya untuk meminimalisasi insidensi dan keparahan
penyakit bercak ter pada budi daya tanaman sorgum. Teknik budi daya ini meliputi
pemilihan genotipe dan kondisi tanam termasuk kondisi lingkungan. Sorghum (Sorghum bicolor) is the fifth most important cereal crop in the world after rice, wheat, corn, and barley which has many benefits such as material for food, feed, and industry. Sorghum can withstand drought and grows in marginal land, so it is suitable to be developed in Indonesia. Sorghum cultivation faces several problem and disease is one of them. Tar spot disease is one of the diseases in sorghum due to infection with the fungus Phyllachora sp. Comprehensive knowledge on the pathogen ecology and resistance response of sorghum to Phyllachora infection is indispensable for the development of tar spot disease-resistant sorghum. Although this disease has a small impact on yield loss, at advanced levels, it will have an impact on considerable yield loss. A study of the relationship between lignin total content and plant defence to disease is necessary since lignin plays an important role in plant resistance to pathogen infections. The objectives of the research were to (1) gaining knowledge about the morphological and molecular characteristics of the fungus Phyllachora sp. on sorghum plants in Indonesia; (2) determining the incidence and severity of tar spot disease in Bogor, Gunung Kidul, and West Lombok; (3) gaining knowledge about the relationship between lignin content and the resistance of sorghum plants to Phyllachora sp. infection through field evaluation tests; and (4) determining the monolignol composition and expression of lignin biosynthesis genes in sorghum plant leaves as variables that affect sorghum plant resistance to tar spot disease. Identification of fungi is generally carried out through morphological and molecular approaches. Several morphological characteristics of the fungus Phyllachora sp. that can be used for identification are the size and form of the ascospores and ascus, as well as the length and character of the paraphysis. Molecular approaches are very important in identifying Phyllachora, which is an obligate fungus, because it is difficult to grow on fungal culture media. Molecular identification of fungi using the polymerase chain reaction (PCR) method is carried out using fungal universal markers, namely markers that amplify the internal transcribed spacer (ITS) and large subunit (LSU) regions, then followed by sequencing analysis. The resistance response of sorghum plants to Phyllachora infection related to the total lignin content in sorghum plants was carried out through field evaluation tests (bioassay). The mechanism of plant resistance, which is based on changes in lignin composition and biosynthesis, was carried out through analysis of the monolignol composition in sorghum leaves using the Pyrolysis Gas Chromatography Mass Spectrometry (Py-GCMS) method. Meanwhile, analysis of the expression of the gene encoding lignin biosynthesis was carried out using the SbCOMT gene marker using the Real-Time Quantitative PCR (RT-qPCR) method. In addition, observations of several agronomic traits related to biomass (including plant height, stem diameter, panicle length, leaf weight, stem weight, panicle weight, and hundred seed weight) were carried out to study the relationship between lignin content and the agronomic characteristics of each genotype. This is important as a basis for determining the type of sorghum genotype that will be planted according to its function, for example, as a source of food, feed and biomaterial product. This research shows that the observed tar spot disease severity values varied from 32.4% in the susceptible Samurai genotype to 87.9% in the Latu Keta genotype, which was resistant to tar spot disease. The results of the morphological identification of 17 genotypes show different characteristics so that they can be classified into multiple species groupings, with the possibility of them becoming new species. Molecular identification with markers that amplify the ITS and LSU regions, respectively, showed that two isolates and eight isolates were Phyllachora fungi. The results of morphological and molecular characterization show consistency supporting the statement that identification and characterization of fungi using a morphological approach is still the key basis for the identification and characterization of fungi that are verified and equipped with a molecular approach. The results of this study also show a correlation between total lignin content and plant resistance to tar spot disease. The results of monolignol composition analysis were comparable and consistent results with the results of SbCOMT gene expression analysis. Genotype 4183A shows high resistance to tar spot disease. It is hoped that the results of this research will provide benefits, including (1) providing foundational knowledge for phytopathologists in studying further about the fungus Phyllachora sp. causes of tar spot disease in sorghum plants. Another advantege is that (2) it becomes useful knowledge for sorghum plant breeding in developing sorghum plants that are resistant to tar spot disease. The final benefit of this research is (3) recommendations for the use of different sorghum genotypes for various purposes, including food, feed, and industry, along with suggestions for cultivation techniques to minimize the incidence and severity of tar spot disease in sorghum cultivation. This cultivation technique of farming involves planting under certain environmental circumstances as well as choosing genotypes. |
