Abstract: | Jakaba atau jamur keberuntungan abadi merupakan salah satu cendawan yang diduga memiliki manfaat berupa nutrisi bagi tanaman sehingga berpotensi untuk dibudidayakan lebih lanjut. Keberhasilan suatu biakan murni didasarkan pada keberhasilan pembuatan media dan kesesuaian lingkungan terhadap pertumbuhan miselium cendawan. Maka dari itu penting untuk dilakukan identifikasi karakteristik cendawan serta analisis jenis media, keasaman media (pH), dan suhu inkubasi yang sesuai untuk pertumbuhan isolat jakaba BHP01. Perlakuan terdiri atas tiga faktor dengan masing-masing faktor terdiri atas 3 taraf. Faktor macam media terdiri atas Potato Dextrose Agar, Malt Extract Agar, dan Malt Pepton Agar, faktor pH terdiri atas pH 4, pH 6, dan pH 8, serta faktor suhu inkubasi, yaitu 10 ℃, ± 28 ℃, dan 35 ℃. Sehingga terdapat 27 macam kombinasi perlakuan dengan tiga ulangan. Rata-rata diameter isolat jakaba tertinggi terdapat pada kombinasi perlakuan M2P1S2 yaitu media Malt Extract Agar, pH 4, dan suhu inkubasi ± 28 ℃. Hasil analisis statistik menunjukkan bahwa macam media dan pH tidak memberikan pengaruh yang nyata terhadap pertumbuhan diamater isolat jakaba, sedangkan suhu memberikan pengaruh yang nyata. Jakaba or perennial lucky mushroom is one of the fungi that is thought to have benefits in the form of nutrients for plants so that it has the potential for further cultivation. The success of a pure culture is based on the success of making media and the suitability of the environment for fungal mycelium growth. Therefore, it is urgent to identify the characteristics of the fungus and analyze the type of media, media acidity (pH), and incubation temperature suitable for the growth of Jakaba isolates. The treatment consisted of three factors with each factor consisting of 3 level. The media type factor consist Potato Dextrose Agar, Malt Extract Agar, and Malt Peptone Agar, the pH factor consists of pH 4, pH 6, pH 8, and the incubation temperature factor 10 ℃, ± 28 ℃, and 35 ℃. So there are 27 kinds of treatment combinations with three replications. The highest average diameter of Jakaba isolates was found in the M2P1S2 treatment combination, Malt Extract Agar media, pH 4, and incubation temperature ± 28 ℃. The results of statistical analysis showed that the type of media and pH did not have a significant effect on the growth of Jakaba isolate diameter, while temperature had a significant effect. |
