Abstract: Metode spektrofotometri biru merupakan metode baku untuk menganalisis kadar amonium. Metode ini menggunakan hipoklorit sebagai pereaksi, tetapi sifatnya yang tidak stabil dapat menurunkan kepekaan metode. Penelitian ini mempelajari degradasi dan menentukan faktor-faktor yang memengaruhi kestabilan pereaksi hipoklorit-fenol serta pengaruhnya pada analisis amonium secara spektrofotometri. Hipoklorit di kondisi penyimpanan dan jenis botol yang berbeda diukur konsentrasi klorin yang tersedianya melalui titrasi iodometri. Hipoklorit tersebut digunakan sebagai pereaksi untuk mengukur kemiringan kurva kalibrasi dengan spektrofotometer UV-Vis. Konsentrasi klorin yang tersedia menurun seiring waktu di semua sampel. Penurunan terbesar terjadi pada sampel dalam botol kaca transparan dan terpapar sinar matahari, yaitu 77,58% dalam 148 hari. Sebaliknya, penurunan terkecil terjadi pada sampel dalam botol kaca gelap dan di lemari pendingin, yaitu 10,92% dalam 135 hari. Meskipun degradasi hipoklorit terdeteksi, tidak ada korelasi antara stabilitas hipoklorit dan nilai kemiringan kurva kalibrasi amonium.
The indophenol spectrophotometry method is a standard method for analyzing ammonium levels. This method employs hypochlorite as a reagent, but it is unstable and can reduce the sensitivity of this method. This study investigated the degradation and determined factors affecting the stability of the hypochlorite-phenol reagent and its impact on the spectrophotometric analysis of ammonium. The chlorine in hypochlorite under different storage conditions and bottle types was measured by iodometric titration. The hypochlorite was employed to measure the slope of the ammonium calibration curve with a UV-Vis spectrophotometer. The available chlorine decreased over time in all samples. The highest reduction occurred in the sample stored in transparent glass bottles and exposed to sunlight, which was 77.58% in 148 days. Conversely, the least reduction occurred in the samples stored in dark glass bottles and inside a refrigerator, which was 10.92% in 135 days. Despite the observed degradation rates, the hypochlorite stability in this study did not correlate with the slope of the ammonium calibration curve.
