Abstract: | Indonesia memproduksi rumput laut hijau dengan jumlah besar setiap
tahunnya, namun hampir seluruhnya dimanfaatkan untuk dikonsumsi langsung.
Padahal, rumput laut hijau memiliki kandungan protein dan ulvan yang cukup
tinggi yang dapat dimanfaatkan pada berbagai sektor industri. Proses ekstraksi
protein atau ulvan dari rumput laut telah dilaporkan oleh beberapa referensi namun
proses ekstraksi yang dilakukan hanya untuk mengekstrak ulvan atau protein saja.
Metode ekstraksi yang terintegrasi belum banyak dikembangkan, padahal metode
ini dapat menekan biaya dan energi yang digunakan. Selain itu, metode ekstraksi
yang ada biasanya bergantung pada penggunaan banyak pelarut kimia untuk
melarutkan protein atau ulvan dari dalam dinding sel. Oleh karena itu, penelitian
ini bertujuan mendesain proses ekstraksi protein dan ulvan dari rumput laut Ulva
sp. secara terintegrasi. Ultrasound-assisted extraction (UAE) digunakan sebagai
metode ekstraksi karena dikenal sebagai green extraction technology. Penelitian ini
menggunakan 5 g bubuk Ulva sp. kering yang diekstrak dengan dua desain. Desain
pertama (F1) mengekstrak Ulva sp. kering dengan UAE pada suhu rendah, dimana
lama ekstraksi divariasikan 10, 15, dan 20 menit. Ulvan dan protein di dalam
ekstrak kemudian dipisahkan dengan metode separasi anionik. Perlakuan tanpa
separasi anionik digunakan sebagai pembanding. Desain kedua (F2) mengekstrak
Ulva sp. kering dengan UAE pada suhu tinggi yang divariasikan waktunya selama
15, 30, dan 45 menit. Desain proses terbaik adalah F1 dengan waktu sonikasi 15
menit, menggunakan separasi anionik yang menghasilkan menghasilkan rendemen
protein terekstrak sebesar 75,84%, lebih tinggi dari rendemen protein terekstrak
dari penelitian sebelumnya. Sedangkan rendemen akhir ulvan hanya berada pada
kisaran 7% bk yang berada di bawah rata-rata ekstraksi pada penelitian yang telah
dilakukan sebelumnya. Indonesia produces large amounts of green seaweed yearly, but almost all of it is used for direct consumption. Green seaweed has a high protein and ulvan content that can be utilized in various industrial sectors. The process of extracting protein or ulvan from seaweed has been reported by several references, but the extraction process is carried out only to extract ulvan or protein alone. An integrated extraction method has not been developed much, even though this method can reduce the cost and energy used. In addition, existing extraction methods usually rely on using many chemical solvents to dissolve proteins or ulvan from within the cell wall. Therefore, this study aims to design an integrated process of protein and ulvan extraction from Ulva sp. seaweed. Ultrasound-assisted extraction (UAE) was used as the extraction method, known as a green extraction technology. This study used 5 g of dried Ulva sp. powder which was extracted by two designs. The first design (F1) extracted dried Ulva sp. with UAE at low temperature, where the extraction time was varied by 10, 15, and 20 minutes. Ulvan and protein in the extract were then separated by anionic separation method. The treatment without anionic separation was used as a comparison. The second design (F2) extracted dried Ulva sp. with UAE at high temperature and varied the time for 15, 30, and 45 minutes. The best process design was F1 with a sonication time of 15 minutes, using anionic separation which resulted in an extracted protein yield of 75.84%, higher than the extracted protein yield from previous studies. The final yield of ulvan is only in the range of 7% DW which is below the average extraction in previous studies. |
