Semak Daun Island is one of the small islands in the southern part of the Kepulauan Seribu Marine National Park, surrounded by seagrass ecosystems. Seagrass ecosystems are a place to find food, shelter, and nursery areas for fish communities. This study aims to analyze the ichthyofaunal biodiversity of the seagrass ecosystem of Semak Daun Island, Seribu Islands Marine National Park, during the rainy season, transition 1, and transition 2 in 2018-2019 and the relationship between fish composition and physicochemical parameters of the waters. Fish sampling were taken using fishing gear of seine nets in the seagrass ecosystem waters of Semak Daun Island was carried out for five months, namely September, November, December 2018, and February and March 2019. Data analysis used included ecological index analysis, analysis of variance, one-way analysis of similarities, and canonical correspondence analysis. During the study, 51 fish species were caught from 24 families, mainly at the juvenile stage. The composition of the fish assemblage differs temporally (season). Canonical correspondence analysis showed that Semak Daun Island fish assemblages positively correlated with seagrass coverage and dissolved oxygen parameters. The ichthyofauna diversity of Semak Island’s seagrass ecosystem is classified as moderated.
